Doxorubicin-induced delayed-onset subclinical cardiotoxicity in mice

Summary: Research suggests that low doses of doxorubicin may lead to cardiotoxicity and subsequent cardiomyopathy. Experiments on mice showed that higher cumulative doses of DOX were associated with decreased cardiac function and increased cardiomyocyte apoptosis.

Summary: Trpc6 deficiency can prevent doxorubicin-induced cardiac damage, especially in male mice. Female mice are less susceptible to doxorubicin-induced cardiac damage and functional changes but Trpc6-deficient females show improved vacuolation under doxorubicin treatment. Sex differences in body weight and expression of Trpc1, Trpc3, and Rcan1 were observed in response to doxorubicin in both wild-type and Trpc6-deficient mice.

Summary: The study revealed that TRIM25 plays a protective role in DOX-induced cardiotoxicity by mitigating endoplasmic reticulum stress and apoptosis. TRIM25 affects the stability and ubiquitination of p85 alpha, leading to increased nuclear translocation of XBP-1s and alleviation of ER stress. These findings suggest that TRIM25 may have therapeutic potential for DOX-induced cardiotoxicity.
