This study characterized the immunogenicity of CLDN6 in ovarian cancer patients and identified specific TCR genes from CD8(+) and CD4(+) T cells, which are important for the treatment of ovarian and other solid tumors expressing CLDN6.

Targeting tumor lineage plasticity in hepatocellular carcinoma using an anti-CLDN6 antibody-drug conjugate

Summary: Tumor lineage plasticity is increasingly recognized as a key mechanism of therapeutic resistance, where Claudin6 (CLDN6) was found to play a role in inducing a phenotypic shift in hepatocellular carcinoma (HCC) cells, making them more difficult to treat. Activation of the CLDN6/TJP2/YAP1 axis and Hippo signaling pathway may enhance tumor lineage plasticity and induce changes in cellular identity. Furthermore, a novel anti-CLDN6 monoclonal antibody conjugated with a cytotoxic agent showed potent antitumor efficacy in HCC treatment.

Summary: The folliculo-stellate (FS) cells in the anterior pituitary glands form follicular lumens sealed by tight junctions (TJs). Occludin and claudin-9 are major components of TJs in FS cells. Claudin-9 expression increases postnatally, showing developmental regulation, and other proteins like angulin-1, angulin-2, and tricellulin are localized at tricellular contacts of FS cells.

Aberrant Claudin-6-Adhesion Signaling Promotes Endometrial Cancer Progression via Estrogen Receptor α

Summary: Cell adhesion proteins, including the CLDN family, play a crucial role in maintaining tissue integrity and regulating cellular events. Aberrant expression of CLDN6 is associated with accelerated proliferation and migration in endometrial cancer cells, promoting tumor growth and invasion. The CLDN6/SFK/PI3K/AKT and SGK signaling pathways activate ERα in an ERα-independent manner, contributing to tumor progression. Moreover, CLDN6 can also regulate gene expression independently of ERα.
