Origin, specification and differentiation of a rare supporting-like lineage in the developing mouse gonad

Chloe Mayere et al.

Summary: This study investigated an uncharacterized population of supporting-like cells (SLCs) in the developing mouse gonads, focusing on their origin, specification, and subsequent sex-specific differentiation. The SLC lineage, closely related to the coelomic epithelium, was specified as early as E10.5, making it the first somatic lineage in the bipotential gonad. SLCs progressively acquired a more Sertoli- or pregranulosa-like identity and contributed to the formation of the rete testis and rete ovarii. WNT4 was found to be a crucial regulator of the SLC lineage and necessary for normal development of the rete testis.

Summary: This review examines the anatomy, endocrine control, and development of the mammalian rete testis, focusing on recent findings on its molecular regulation, identifying gaps in our knowledge, and identifying areas for future research. Most recently, modern molecular approaches have revealed the transcriptional signature of rete cell precursors, pointing to at least a partial common origin with Sertoli cells. Application of technologies such as single-cell RNA-seq will clarify the origin and developmental trajectory of this essential component of the male reproductive tract.

Summary: The rete testis is an essential component of the male reproductive tract, developing in close association with Sertoli cells. However, there is limited understanding of its molecular markers and regulatory mechanisms, presenting gaps in current knowledge that could be addressed through the application of modern molecular technologies such as single-cell RNA-seq for future research.
