Molecular Basis of Small-Molecule Binding to alpha-Synuclein

Paul Robustelli et al.

Summary: This study used molecular dynamics simulations to investigate the binding of a small-molecule drug to an intrinsically disordered protein associated with Parkinson's disease. The simulations revealed a dynamic shuttling mechanism of certain charge-charge and pi-stacking interactions during the binding process. Subsequent experiments confirmed the potential for designing small molecules that effectively bind with disordered proteins.

Amyloid Oligomers: A Joint Experimental/Computational Perspective on Alzheimer's Disease, Parkinson's Disease, Type II Diabetes, and Amyotrophic Lateral Sclerosis

Phuong H. Nguyen et al.

Summary: Protein misfolding and aggregation are common in amyloidogenic diseases, and understanding how amyloid species become toxic is crucial for treatment development. Experiments using computer modeling, in vitro and in vivo studies, as well as pharmacology, have provided valuable insights into the accumulation and deposition of amyloid species, contributing to research on diseases like Alzheimer's, Parkinson's, diabetes, and ALS.

Sequence Determines the Switch in the Fibril Forming Regions in the Low-Complexity FUS Protein and Its Variants

Abhinaw Kumar et al.

Summary: Residues spanning distinct regions of the low-complexity domain of the FUS-LC RNA-binding protein form fibril structures with different core morphologies, S-bend for FUS-LC and U-bend for FUS-LC-C. The formation of core-1 is favored compared to core-2, and computer simulations suggest that the two fibril cores do not coexist.

α-Synuclein: An All-Inclusive Trip Around its Structure, Influencing Factors and Applied Techniques

Nicolo Bisi et al.

Summary: Alpha-synuclein is a highly expressed and conserved protein found in neuronal presynaptic terminals, and its misfolding and aggregation are associated with several neurodegenerative diseases. Studies have shown that the structure of alpha-synuclein can be influenced by various factors, including mutations, biological environment, and interactions with exogenous compounds.
