METTL3-mediated m6A modification of ATG7 regulates autophagy-GATA4 axis to promote cellular senescence and osteoarthritis progression

Xiang Chen et al.

Summary: This study revealed the significant increase of senescent FLSs with OA progression, impacting autophagy and SASP expression. METTL3 negatively regulated autophagy-related genes, promoting FLS senescence. Inhibiting METTL3 could attenuate FLS senescence, offering a potential therapeutic strategy for OA.

From Development to Aging: The Path to Cellular Senescence

Alessandra Sacco et al.

Summary: Senescence is a cellular state induced by internal or external stimuli, resulting in cell cycle arrest, morphological changes, and dysfunctions in mitochondrial and lysosomal functionality. Recent evidence suggests that senescent cells in vivo are a heterogeneous cell population due to different activated pathways and distinct microenvironmental contexts. Future research will focus on exploring the heterogeneity of senescent cells to map cell progression and study the therapeutic advantages of senolytic drugs.

Biological and epigenetic alterations of mitochondria involved in cellular replicative and hydrogen peroxide-induced premature senescence of human embryonic lung fibroblasts

Yan Wang et al.

Summary: The study revealed that in the senescence process of human embryonic lung fibroblasts, mitochondrial quantity decreased, mtDNA copy number increased, methyltransferases activity heightened, and acute ROS accumulation triggered premature senescence. In persistent premature senescence, ATP levels and mt-5-mC were higher than in replicative senescence, while 8-OHdG levels were lower. Alterations in mitochondrial biological functions and mtDNA features may serve as novel biomarkers for cellular senescence.

Post-translational modification of RNA m6A demethylase ALKBH5 regulates ROS-induced DNA damage response

Fang Yu et al.

Summary: The RNA demethylase ALKBH5 serves as a key regulator in protecting cells from DNA damage and apoptosis during ROS-induced stress. ROS induces global mRNA N-6-methyladenosine levels by modulating ALKBH5 post-translational modifications, leading to the rapid induction of genes involved in DNA damage repair. Additionally, ROS activates the ERK/JNK/ALKBH5-PTMs/m(6)A axis in hematopoietic stem/progenitor cells in mice, indicating a physiological role of this pathway in maintaining genome stability.

Inflammation, epigenetics, and metabolism converge to cell senescence and ageing: the regulation and intervention

Xudong Zhu et al.

Summary: Significant progress has been made in ageing research, with a focus on epigenetic dynamics and immunometabolic reprogramming in relation to cell senescence. These hotspots not only shape cell identity, but also play a role in cell fate determination. Potential therapeutic interventions targeting cell senescence and regulatory mechanisms using state-of-the-art techniques are discussed.

N6-methyladenosine dynamics in neurodevelopment and aging, and its potential role in Alzheimer's disease

Andrew M. Shafik et al.

Summary: The study reveals that m(6)A modification exhibits temporal and spatial dynamics in neurodevelopment and aging, with tissue-specific methylation associated with mRNA expression and tissue-specific processes. Aging leads to an increase in m(6)A sites, with a negative effect on mRNA expression, and Alzheimer's disease-related genes show decreased m(6)A methylation correlated with reduced protein levels.
