Molecular identification and pathogenicity of Rhizoctonia solani and Pythium spp. associated with damping-off disease on baby leafy vegetables in Greece

George T. Tziros et al.

Summary: In recent years, the cultivation of leafy vegetables as baby leaves has gained recognition in the global market and has attracted the interest of consumers. This study conducted a comprehensive analysis of specific Rhizoctonia solani AGs and Pythium spp. derived from baby leafy vegetables exhibiting damping-off symptoms in Greece. The results showed variations in pathogenicity among the different AGs of R. solani and identified three Pythium spp. The findings highlight the importance of understanding these pathogens in order to prevent and manage damping-off symptoms in baby leaf vegetables.

CRISPR-Cas9 approach confirms Calcineurin-responsive zinc finger 1 (Crz1) transcription factor as a promising therapeutic target in echinocandin-resistant Candida glabrata

Andres Ceballos-Garzon et al.

Summary: Invasive fungal infections are difficult to treat due to limited antifungal drugs and antifungal resistance. This study suggests that the transcription factor Crz1, which is absent in human cells, could be a potential therapeutic target for fungal infections. Inhibition of Crz1 may restore susceptibility to antifungal drugs and affect various aspects of fungal pathogenicity.

RS_CRZ1, a C2H2-Type Transcription Factor Is Required for Pathogenesis of Rhizoctonia solani AG1-IA in Tomato

Srayan Ghosh et al.

Summary: The study successfully employed HIGS and dsRNA technologies to downregulate pathogenic genes of R. solani in tomato, with significant reduction in disease symptoms and pathogen colonization depth observed upon silencing of the RS_CRZ1 gene. Additionally, coregulated genes with RS_CRZ1 were identified during the pathogenicity process, highlighting the importance of RS_CRZ1 in managing the hostile environment encountered during host colonization and the potential of gene silencing approaches for functional characterization of pathogenicity determinants of R. solani.

Improving evaluation of potato resistance to Rhizoctonia solani infection by optimizing inoculum-based method combined with toxin-based assay

Xiaoyu Zhang et al.

Summary: Rhizoctonia solani causes stem canker in potatoes and affects production significantly. Testing potato resistance to R. solani using pathogen inoculum can be time consuming. A study showed that inoculation with 2, 3, or 4 grams of R. solani AG2-1 effectively distinguished resistance levels in potato cultivars/lines.

Pattern-recognition receptors are required for NLR-mediated plant immunity

Minhang Yuan et al.

Summary: The plant immune system consists of pattern-triggered immunity (PTI) and effector-triggered immunity (ETI), which are activated by different mechanisms and involve distinct signaling cascades. PTI is triggered by microbial patterns via cell surface receptors, while ETI is activated by pathogen effector proteins via intracellular receptors.

Spray-induced gene silencing for disease control is dependent on the efficiency of pathogen RNA uptake

Lulu Qiao et al.

Summary: Recent discoveries show that fungi can uptake environmental RNA to silence fungal genes through RNA interference, leading to the development of Spray-Induced Gene Silencing (SIGS) for plant disease management. The efficiency of RNA uptake varies across eukaryotic microbe species and cell types, which plays a critical role in determining the success of SIGS for plant disease management.

Modulation of fungal virulence through CRZ1 regulated F-BAR-dependent actin remodeling and endocytosis in chickpea infecting phytopathogen Ascochyta rabiei

Manisha Sinha et al.

Summary: ArF-BAR protein in the filamentous fungus Ascochyta rabiei regulates endocytosis and actin dynamics in hyphal growth, playing a crucial role in fungal virulence on the host plant. Its functional significance is highlighted by its regulation through a stress-induced transcription factor and involvement in effector secretion and actin cytoskeleton regulation. This study sheds light on the importance of F-BAR domain proteins in fungal pathophysiology, especially in the context of plant infection.

Functional validation of pathogenicity genes in rice sheath blight pathogen Rhizoctonia solani by a novel host-induced gene silencing system

Mei Zhao et al.

Summary: A newly established TRV-HIGS system was used to validate pathogenicity genes in the rice sheath blight pathogen R. solani, with 29 out of 33 candidate genes showing significant reduction in necrosis caused by GD-118 infection. Stable rice transformants harboring double-stranded RNA constructs for one positive gene, trehalose-6-phosphate phosphatase (Rstps2), exhibited reduced gene expression, virulence, and trehalose accumulation in GD-118, indicating the potential for controlling the disease through gene silencing techniques.

Calcium signaling is involved in diverse cellular processes in fungi

Avishek Roy et al.

Summary: Calcium (Ca2+) is a universal signalling molecule crucial for various cellular processes. Maintaining a low resting level of intracellular free Ca2+ concentration is essential for cell function, as high concentrations of Ca2+ can be harmful. Ca2+ signalling plays a significant role in growth, development, reproduction, stress tolerance, and virulence in fungi.
