Dihydronicotinamide Riboside Is a Potent NAD+ Precursor Promoting a Pro-Inflammatory Phenotype in Macrophages

Summary: Nicotinamide adenine dinucleotide (NAD) metabolism plays a crucial role in immune function regulation, but the complete understanding of how NAD and its metabolites, precursors, and enzymes work together in immune function and inflammatory diseases is still lacking. This study found that supplementation of NAD precursor NRH significantly increased NAD(+) levels in macrophages and activated a pro-inflammatory phenotype, inducing the expression of multiple cytokines, chemokines, and enzymes. Furthermore, NRH enhanced the effect of lipopolysaccharide on macrophage activation and cytokine gene expression.
