Differential Effects of Platelet Factor 4 (CXCL4) and Its Non-Allelic Variant (CXCL4L1) on Cultured Human Vascular Smooth Muscle Cells

Dawid M. Kaczor et al.

Summary: Platelet factor 4 (CXCL4) is a chemokine stored in platelets that modulates the function of vascular smooth muscle cells (VSMCs). Variant CXCL4L1, despite minor differences, has distinct effects on VSMCs compared to CXCL4. Both CXCL4 and CXCL4L1 regulate gene expression, but only CXCL4 increases VSMC proliferation and calcification.

Platelet Extracellular Vesicles: Beyond the Blood

Florian Puhm et al.

Summary: Extracellular vesicles (EVs) are a means of cell-to-cell communication, and platelets readily generate EVs upon activation, containing diverse cargo that can enter various bodily fluids. The importance of platelet-derived EVs extends beyond hemostasis, highlighting their physiological and pathological significance.

Platelet-rich plasma-derived extracellular vesicles: A superior alternative in regenerative medicine?

Jiuping Wu et al.

Summary: Platelet-rich plasma (PRP) has been used in regenerative medicine for over 30 years due to its promising therapeutic properties. Recent studies have found that extracellular vesicles (EVs) released from activated platelets also play a role in tissue repair, leading to increased research on PRP-derived EVs (PRP-EVs) for tissue regeneration as a superior cell-free therapeutic strategy.

Platelet-derived extracellular vesicles to target plaque inflammation for effective anti-atherosclerotic therapy

Summary: Engineering platelet-derived extracellular vesicles (PEVs) to load MCC950, an NLRP3-inflammasome inhibitor, for atherosclerosis-targeted therapy can significantly reduce plaque formation, lower local inflammation, and inhibit proliferation of macrophages and T cells at the plaque site compared with free drug administration. This strategy indicates the promising potential of PEVs for targeted drug delivery in atherosclerosis treatment.

Summary: The application of platelet-rich plasma (PRP) in oral and maxillofacial surgery has been thoroughly studied, with different types of PRP showing varying efficacy outcomes depending on their composition and studied application. P-PRP (without leukocytes) has been more consistent in achieving beneficial effects in alveolar ridge preservation, management of post-extraction complications, bone augmentation, and temporomandibular joint disorders, highlighting the importance of composition and methodology in evaluating PRP efficacy.

Summary: Cells communicate with each other via extracellular vesicles (EVs). These EVs carry various types of cargo and can have different effects on recipient cells. They play important roles in atherosclerosis and COVID-19-related thrombosis, and have potential as signaling mediators, biomarkers, and therapeutic agents. More research is needed to fully understand the mechanisms of EVs in normal and pathological conditions.

Platelet-Rich Plasma-Derived Exosomal USP15 Promotes Cutaneous Wound Healing via Deubiquitinating EIF4A1

Yan Xu et al.

Summary: This study found that PRP-Exos effectively promoted the in vitro proliferation, migration, and wound healing activity of HaCaT cells. USP15 was identified as a key mediator in promoting tissue repair both in vitro and in vivo, enhancing the functional properties of HaCaT cells by promoting EIF4A1 deubiquitination. Thus, PRP-Exos and USP15 are promising tools for enhancing wound healing through promoting epithelial regeneration.
