Renal cell carcinoma therapy: Current and new drug candidates

Olivia Pontes et al.

Summary: Renal cell carcinoma is the most common and lethal tumor of the urological system, with various treatment options available including surgery and targeted therapies. However, the development of resistance and limited durable responses highlight the need for new anticancer candidates with improved selectivity and efficacy. Recent research has focused on novel natural and synthetic compounds for treating RCC.

Inositol depletion regulates phospholipid metabolism and activates stress signaling in HEK293T cells

Mahmoud Suliman et al.

Summary: Inositol plays a significant role in cellular function and signaling. Its deficiency can cause cell death and abnormalities in inositol metabolism are associated with human disorders. This study provides a comprehensive understanding of the effects of inositol deprivation on phospholipid metabolism and gene expression in human cells, revealing the essential role of inositol in maintaining cell viability and regulating cell signaling and metabolism.

Reactive oxygen species (ROS) in cancer pathogenesis and therapy: An update on the role of ROS in anticancer action of benzophenanthridine alkaloids

Abdul Q. Khan et al.

Summary: Reactive oxygen species (ROS) play a crucial role in the growth, progression, and stemness of cancer cells, maintaining a delicate balance with antioxidants to promote cancer pathogenesis. Natural products are vital in attenuating the adverse effects of cancer therapies, with most available drugs derived from natural resources. Benzophenanthridine alkaloids, such as sanguinarine, exhibit pharmacological properties and are being studied for the treatment of various diseases, including cancer, by inducing ROS-mediated anticancer actions.

Metabolomic Analysis to Elucidate Mechanisms of Sunitinib Resistance in Renal Cell Carcinoma

Tomonori Sato et al.

Summary: Metabolomics analysis can identify potential therapeutic targets for treatment resistance in cancer, such as sunitinib resistance in renal cell carcinoma (RCC), through changes in metabolites. This study found that alterations in energy metabolism, glycolysis upregulation, and antioxidant activity are associated with sunitinib resistance in RCC cells.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

Crizotinib and Sunitinib Induce Hepatotoxicity and Mitochondrial Apoptosis in L02 Cells via ROS and Nrf2 Signaling Pathway

Lin Guo et al.

Summary: The study investigated the mechanisms of crizotinib and sunitinib-induced hepatotoxicity and potential mitigation strategies through ROS and Nrf2 signaling. Both drugs activated oxidative stress and triggered hepatocyte mitochondrial apoptosis and necrosis in liver cells. ROS scavenger NAC and Nrf2 agonist tBHQ were found to alleviate cell damage and mitochondrial apoptosis during crizotinib- and sunitinib-induced hepatotoxicity.

Molecular and Functional Analysis of Sunitinib-Resistance Induction in Human Renal Cell Carcinoma Cells

Magdalena Rausch et al.

Summary: Resistance against sunitinib in clear cell renal cell carcinoma (ccRCC) involves various molecular aberrations, leading to clinical complications. Understanding these aberrations and developing an optimized multidrug combination is crucial to surpassing resistance mechanisms and improving treatment outcomes. Research on resistance mechanisms revealed morphometric and molecular changes, as well as the presence of N-desethyl sunitinib and lysosomal sequestration of the drug, highlighting the importance of studying drug responses in laboratory settings.
