Neurofilament light chain in blood as a diagnostic and predictive biomarker for multiple sclerosis: A systematic review and meta-analysis

Liangxia Ning et al.

Summary: This meta-analysis assessed the diagnosis and predictive value of blood neurofilament light chain (NfL) in multiple sclerosis (MS). The results suggest that blood NfL may be a useful biomarker in diagnosing MS, distinguishing MS subtypes, and predicting disease worsening in the future.

Oligoclonal IgM bands in the cerebrospinal fluid of patients with relapsing MS to inform long-term MS disability

Rocco Capuano et al.

Summary: This study investigated the role of cerebrospinal fluid (CSF) markers in predicting long-term multiple sclerosis outcomes, with the presence of oligoclonal IgM bands (OCMB) associated with unfavorable long-term outcomes. These findings suggest that OCMB should be determined in relapsing MS patients to inform long-term prognosis.

The role of astrocyte-mediated plasticity in neural circuit development and function

Nelson A. Perez-Catalan et al.

Summary: Neural plasticity is crucial for shaping circuit function during nervous system development, and is regulated by astrocytes, the most abundant glial cell type in the human nervous system. Recent technological advancements have highlighted the important role of astrocytes in regulating circuit assembly and function.

First line treatment failure: Predictive factors in a cohort of 863 Relapsing Remitting MS patients

Xavier Ayrignac et al.

Summary: This study found that patients younger than 26 years old, those with an EDSS score greater than 2, and those with more than 2 relapses in the previous year are more likely to experience treatment failure with first line therapy for RR-MS. Factors significantly associated with treatment failure also include EDSS progression and age at treatment start.

The Increasing Role of Kappa Free Light Chains in the Diagnosis of Multiple Sclerosis

Franz Felix Konen et al.

Summary: Free light chains (FLC), particularly the kappa isoform (KFLC), show high diagnostic sensitivity and specificity in detecting intrathecal inflammation in central nervous system diseases. The measurement of KFLC is fully automated, stable against most influencing factors, and represents a promising diagnostic approach for neuroinflammatory diseases. Further multicenter studies are needed to establish the diagnostic sensitivity and specificity of KFLC, using standardized cut-off values and non-linear formulas.

Manipulating oligodendrocyte intrinsic regeneration mechanism to promote remyelination

Fabien Biname et al.

Summary: In demyelinated lesions, the failure of myelin repair is largely attributed to the accumulation of inhibitory cues and opposing factors, leading to inefficient remyelination. The complex interaction between signaling pathways controlling oligodendrocyte migration or differentiation may pave the way for novel therapeutic approaches by modulating the global response to inhibitory cues.

Inflammation in multiple sclerosis

Stefanie Haase et al.

Summary: Multiple sclerosis (MS) is a chronic inflammatory disease of the central nervous system (CNS). Its underlying pathomechanisms involve autoimmune responses towards CNS self-antigens, including autoreactive T cells, B cells, and myeloid cells. Current therapies aim to shift the immune cell repertoire from pro-inflammatory to anti-inflammatory, involving regulatory T and B cells, and anti-inflammatory macrophages.
