Identification of Altered Blood MicroRNAs and Plasma Proteins in a Rat Model of Parkinson's Disease

Sanjeev Kumar Yadav et al.

Summary: This study aimed to characterize potential biomarkers of Parkinson's disease using miRNomics, proteomics, and bioinformatics approaches. Rats exposed to rotenone were used as a model, and a set of miRNAs and proteins significantly deregulated in blood samples were identified. Pathway analysis revealed multiple pathways leading to Parkinson's disease.

Genome-wide study of DNA methylation shows alterations in metabolic, inflammatory, and cholesterol pathways in ALS

Paul J. Hop et al.

Summary: This study conducted a blood-based epigenome-wide association study meta-analysis and identified differentially methylated positions (DMPs) and genes associated with ALS. The study also found independent associations between ALS and high-density lipoprotein cholesterol, body mass index, white blood cell proportions, and alcohol intake, with cholesterol biosynthesis potentially causally related to ALS. Additionally, DNA methylation at several DMPs and blood cell proportion estimates were associated with survival rate in patients.

Co-Expression Analysis of microRNAs and Proteins in Brain of Alzheimer's Disease Patients

Callum N. Watson et al.

Summary: Alzheimer's disease is the most common form of dementia globally, however, its cause is still unknown, hindering the development of effective treatments. Recent studies have found that MicroRNAs play an important role in Alzheimer's disease as regulators of gene expression, and could serve as biomarkers and potential therapeutic targets. This study identified six common pathways regulated by MicroRNAs and proteins, including one pathway that is activated in the early phase, providing opportunities for early intervention.

Genome-Wide Methylation Profiling in the Thalamus of Scrapie Sheep

Adelaida Hernaiz et al.

Summary: This study found DNA methylation alterations in the thalamus of sheep infected with scrapie, a form of transmissible spongiform encephalopathy. Hypomethylated regions were associated with genes involved in transmembrane transport and cell adhesion, while hypermethylated regions were related to genes involved in intracellular signal transduction. Furthermore, differentially expressed genes and genes highly expressed in specific types of CNS cells contained differentially methylated regions.

Interactions of amyloid coaggregates with biomolecules and its relevance to neurodegeneration

Kazuma Murakami et al.

Summary: The aggregation of amyloidogenic proteins is a pathological hallmark of various neurodegenerative diseases, but the molecular mechanisms behind these diseases are not yet fully understood. Different amyloid species can coexist in the same brain and even occur in individuals without disease. Advances in research related to biomolecular condensates and structural analysis have provided insights into the cross-seeding and cross-inhibition of amyloidosis. The potential clinical applications in central nervous system therapy are also highlighted.

Profiling of Serum Exosome MiRNA Reveals the Potential of a MiRNA Panel as Diagnostic Biomarker for Alzheimer's Disease

Zhiwu Dong et al.

Summary: Alzheimer's disease (AD) is the most common neurodegenerative disease in older adults, and identifying peripheral blood-based biomarkers for AD diagnosis is urgently needed. By analyzing the miRNA profile in human serum exosomes, potential biomarkers, such as miR-30b-5p, miR-22-3p, and miR-378a-3p, were found to distinguish AD from healthy controls (HC). The combination of these three miRNAs showed improved diagnostic capability, indicating their potential as biomarkers for AD diagnosis.

Small Non-coding RNAs Are Dysregulated in Huntington's Disease Transgenic Mice Independently of the Therapeutic Effects of an Environmental Intervention

Celine Dubois et al.

Summary: Huntington's disease is a neurodegenerative disorder caused by a gene mutation that affects small non-coding RNAs such as miRNAs, and environmental enrichment can improve related symptoms; however, the mechanisms behind the therapeutic effects are still unclear, with the study showing that HD mainly affects miRNAs and has a minor effect on other sncRNA populations, suggesting that the therapeutic effects of environmental enrichment may be exerted through other molecular mechanisms.

Summary: ALS is a terminal neurodegenerative disease where pathology may spread in an organized and prion-like manner through extracellular vesicles. The study found differences in size of EVs between ALS patients and healthy controls in spinal cord and serum, as well as altered miRNA levels in ALS frontal cortex, spinal cord, and serum. The dysregulated miRNAs were associated with pathways commonly altered in neurodegeneration, offering potential insights into pathogenic factors of ALS.

Summary: Parkinson's disease (PD) is the second most common age-related neurodegenerative disease, diagnosed at advanced stages by motor deficits. Aberrant epigenetic modifications are detectable in blood and our study found differentially expressed genes (DEGs) and methylated regions (DMRs) in PD patients compared to healthy controls. Identified patterns of altered disease-specific DNA methylation and associated gene expression in whole blood provide a foundation for potential PD biomarkers.

Summary: Cumulative evidence suggests that microRNAs (miRNAs) play a significant role in neurodegenerative diseases, particularly with more pronounced differential expression in the serum of Parkinson's disease patients. The study indicates that miRNA-protein binding may mediate the pathogenic effects of HNF4a, suggesting a complex role for miRNAs in pathology beyond transcriptional dysregulation.
