Extracellular Vesicles as Biomarkers of Acute Graft-vs.-Host Disease After Haploidentical Stem Cell Transplantation and Post-Transplant Cyclophosphamide

Giuseppe Lia et al.

Summary: Even with high-dose PT-Cy, acute and chronic GvHDs remain a clinical challenge. Reliable biomarkers for predicting their onset have yet to be identified. This study found a significant association between the expression profiles of EVs and their miRNA cargo with the onset of aGvHD. The correlation between EV expression profile and cargo with plasma biomarkers was also investigated.

Extracellular Vesicles Derived from Mesenchymal Stromal Cells Delivered during Hypothermic Oxygenated Machine Perfusion Repair Ischemic/Reperfusion Damage of Kidneys from Extended Criteria Donors

Teresa Rampino et al.

Summary: This study explores the use of hypothermic oxygenated perfusion (HOPE) combined with extracellular vesicles (EV) derived from mesenchymal stromal cells as a new strategy for organ preservation. The results show that EV delivery during HOPE can effectively reduce ischemic damage, prevent reoxygenation-dependent injury, and improve transplant outcomes. This study is significant in increasing the donor pool and improving transplantation success.

Mesenchymal Stem Cell-Derived Extracellular Vesicles Attenuate Mitochondrial Damage and Inflammation by Stabilizing Mitochondrial DNA

Meng Zhao et al.

Summary: The study demonstrates that MSC-EVs can alleviate mtDNA damage and inflammation after acute kidney injury, with their effects partially dependent on the TFAM signaling pathway. This suggests that MSC-EVs are promising nanotherapeutics for diseases characterized by mitochondrial damage, highlighting the essential role of TFAM in maintaining their regenerative capacity.

Ex-vivo Kidney Machine Perfusion: Therapeutic Potential

Ruta Zulpaite et al.

Summary: Kidney transplantation is the gold standard treatment for patients with end-stage kidney disease, but the quality of donor organs poses limitations. Ex-vivo machine perfusion technology has great potential in maintaining kidney viability and optimizing organ treatment, serving as an important component of novel techniques in transplant surgery.

Extracellular ATP and Adenosine in Cancer Pathogenesis and Treatment

Anna M. Chiarella et al.

Summary: ATP hydrolysis and downstream signaling pathways play a crucial role in tumor progression and metastasis in the extracellular space. The complexity and interdependence of various cell types in the extracellular space are increasingly recognized, leading to attention on therapeutic strategies targeting extracellular adenosine metabolism and signaling in the development and progression of malignancies. This review provides a summary of the molecular and physiological roles of extracellular ATP and adenosine in normal and disease states, as well as their potential therapeutic applications.
