Molecular crowding facilitates bundling of IMPDH polymers and cytoophidium formation

Chia-Chun Chang et al.

Summary: The cytoophidium is a unique membraneless compartment composed of filamentous protein polymers. This study reveals that human IMPDH2 polymers can form cytoophidium-like aggregates in the presence of macromolecular crowders. Molecular crowding and RNA synthesis inhibition affect cytoophidium assembly. In addition to IMPDH, other proteins can also form cytoophidium. Furthermore, the cytoophidium can prolong the half-life of IMPDH.

Cytoophidia coupling adipose architecture and metabolism

Jingnan Liu et al.

Summary: The metabolic enzyme CTP synthase (CTPS) forms filamentous structures called cytoophidia along the adipocyte cortex, influencing integrin distribution and collagen deposition, ultimately regulating tissue architecture and metabolism in Drosophila adipose tissue.

Hypoosmolality impedes cytoophidium integrity during nitrogen starvation

Yi-Lan Li et al.

Summary: This research compares the differences in cytoophidia between budding yeast and fission yeast, revealing the impact of environmental factors on cytoophidium integrity. After adapting to the optimal medium, cytoophidium fragmentation and morphological changes were observed, indicating the role of cytoophidia in responding to environmental stress.

Polarised maintenance of cytoophidia in Drosophila follicle epithelia

Qiao-Qi Wang et al.

Summary: CTPS can form cytoophidia on the basolateral side of Drosophila follicle cells, with stability and distribution influenced by apical polarity regulators rather than basolateral polarity regulators. This suggests that cytoophidia maintenance in Drosophila follicle epithelia is primarily achieved through the polarised distribution on the basolateral side.

Structural basis for ligand binding modes of CTP synthase

Xian Zhou et al.

Summary: CTPS activity is regulated by the binding of nucleotides and glutamine, with GTP directly blocking ammonia leakage and stabilizing the ammonia tunnel. Cryo-electron microscopy revealed the binding modes of substrates and analogs in Drosophila CTPS, providing insights into allosteric regulation and CTP synthesis mechanisms.
