Circadian rhythms in neurodegenerative disorders

Malik Nassan et al.

Summary: The circadian system governs physiological processes like sleep, alertness, and cognition, and disruptions in circadian homeostasis can have detrimental effects on human health, especially in neurodegenerative disorders. There is a bidirectional relationship between circadian function and neurodegeneration, highlighting the importance of studying circadian disruption in these disorders. Research in this field could lead to novel circadian-based therapeutic interventions for disabling conditions like Parkinson's, Alzheimer's, and other neurodegenerative diseases.

ADARs act as potent regulators of circular transcriptome in cancer

Haoqing Shen et al.

Summary: This study demonstrates that ADARs play a significant role in regulating the circular transcriptome. ADARs can stabilize or destabilize circRNA secondary structures through editing, and also enhance the binding of RNA-binding proteins. These ADARs-regulated circRNAs are widely expressed in various cancers and have functional relevance to tumorigenesis.

The metastatic spread of breast cancer accelerates during sleep

Zoi Diamantopoulou et al.

Summary: The generation of circulating tumor cells (CTCs) with metastatic ability is concentrated during the rest phase, and CTCs generated during the active phase lack metastatic ability. Mechanistically, the upregulation of mitotic genes during the rest phase enables CTCs' metastasis proficiency. Circadian rhythm hormones such as melatonin, testosterone and glucocorticoids dictate the generation dynamics of CTCs, and insulin promotes tumor cell proliferation in a time-dependent manner.

Microglial circadian clock regulation of microglial structural complexity, dendritic spine density and inflammatory response

Hiroshi Nakanishi et al.

Summary: Microglia in the cortex exhibit different shapes and functions during wakefulness and sleep, with their molecular circadian clock playing a key role in regulating these processes. Disruption of the microglial clock system could lead to sleep disturbances, impaired social interaction, and cognitive impairment, potentially contributing to the pathogenesis of neurodegenerative disorders such as Alzheimer's disease. Understanding and targeting the microglial circadian molecular clock may offer insights for developing novel pharmacological interventions for neuropsychiatric and neurodegenerative disorders.

A genome-wide microRNA screen identifies the microRNA-183/96/182 cluster as a modulator of circadian rhythms

Lili Zhou et al.

Summary: This study identified the miRNA cluster miR-183/96/182 as key regulators of circadian rhythms, with miR-96 directly targeting the core circadian clock gene PER2. Knockout of the miR-183/96/182 cluster in mice showed tissue-specific effects on circadian parameters, highlighting the importance of miRNAs as a genome-wide layer of circadian clock regulation.

NPAS4 regulates the transcriptional response of the suprachiasmatic nucleus to light and circadian behavior

Pin Xu et al.

Summary: The suprachiasmatic nucleus (SCN) in mammals is regulated by environmental light and shows responses to light through different peptidergic cell types. The study found that NPAS4 plays a critical role in normal transcriptional responses to light in the SCN, affecting circadian behavior and light-induced gene expression.
