Methods for the identification and characterization of extracellular vesicles in cardiovascular studies: from exosomes to microvesicles

Sean M. Davidson et al.

Summary: This article provides an overview of the techniques and technologies available for the separation and characterization of EVs from different sources. It discusses methods for determining the protein, RNA, and lipid content of EVs. The guidance provided in this document is important for investigating the role of EVs in cardiovascular diseases and highlights key methodological issues to consider.

Associations Among PCSK9 Levels, Atherosclerosis-Derived Extracellular Vesicles, and Their miRNA Content in Adults With Obesity

Chiara Macchi et al.

Summary: This study investigates the interplay among extracellular vesicles (EVs), miRNAs, and proprotein convertase subtilisin/kexin type 9 (PCSK9) in individuals with obesity and a raised cardiovascular risk. The results show that PCSK9 levels are associated with different types of EVs and platelet counts, and are significantly associated with five EV-derived miRNAs. These findings suggest a potential role of PCSK9 in regulating EV-derived miRNAs, especially those related to inflammation and low-density lipoprotein receptor expression.

Platelet-released extracellular vesicles: the effects of thrombin activation

Rosa Suades et al.

Summary: Platelets play essential roles in thrombosis, inflammation, and angiogenesis. Stimulation by thrombin triggers the release of pEVs from platelets, which are rich in procoagulant and proadhesive proteins. Differential proteomic analysis revealed significant changes in protein composition between pEVs from activated platelets and non-activated platelets.

A standarized protocol for evaluation of large extracellular vesicles using the attuneTM NXT system

Leslie Marisol Lugo-Gavidia et al.

Summary: Extracellular vesicles (EVs) are promising biomarkers in several medical areas, but their evaluation using flow cytometry represents a challenge due to their size falling below the detection limit of most standard flow cytometers. The optimization process is crucial to ensure instrument-specific sensitivity.

Arterial Stiffness and Cardiovascular Risk in Hypertension

Pierre Boutouyrie et al.

Summary: Arterial stiffness is a key marker of risk in hypertension, and can be measured at material or structural levels. Studies have shown that structural stiffness predicts outcomes, with elastic arteries being most sensitive to blood pressure and age. Stiffness is considered an index of vascular aging and affects brain, kidneys, and heart function. Reduction of stiffness through antihypertensive treatments and preventive drugs may have additional benefits beyond lowering blood pressure. Newer anti-inflammatory drugs also have effects on stiffness independent of blood pressure.

Role of Microparticles in Cardiovascular Disease Implications for Endothelial Dysfunction, Thrombosis, and Inflammation

Leslie Marisol Lugo-Gavidia et al.

Summary: Microparticles are small cell vesicles derived from the cell membrane, with growing evidence linking them to cardiovascular disease through inflammation, endothelial function, and thrombosis. Research on the direct causal link between microparticle release and their implications on physiological and pathological conditions is ongoing.
