Molecular Aspects of Insulin Aggregation and Various Therapeutic Interventions

In the case of insulin, its aggregation hinders proper glucose level control in diabetic patients. Synthesis and Evaluation of Arylamides with Hydrophobic Side Chains for Insulin Aggregation Inhibition

Summary: By synthesizing a small library of oligopyridylamide foldamers decorated with different hydrophobic side chains, a hit molecule BPAD2 was identified with an IC50 value of 0.9 μM for inhibiting insulin aggregation. Mechanistic analyses showed that BPAD2 mainly inhibited secondary nucleation and elongation processes during aggregation, highlighting the importance of structure-activity relationship studies in understanding protein aggregation at a molecular level.

Prion-derived tetrapeptide stabilizes thermolabile insulin via conformational trapping

Summary: A new tetrapeptide VYYR has been found to effectively inhibit insulin fibrillation and maintain its bioactivity, providing good blood sugar control in mouse models of diabetes. The critical interaction between insulin and IS1 has been elucidated through nuclear magnetic resonance spectroscopy, and a non-canonical dimer-mediated conformational trapping mechanism has been demonstrated for insulin stabilization. This study introduces a class of tetrapeptides for developing thermostable insulin formulations with therapeutic relevance.

Interaction between phloretin and insulin: a spectroscopic study

Summary: The study revealed that phloretin (PHL) has a significant interaction with insulin (INS), forming hydrogen bonds and hydrophobic interactions. Temperature changes affect the interaction, causing a redshift in the emission wavelength of 305 to 317 nm. The thermodynamic parameters show that the interaction between PHL and INS is spontaneous, with enthalpy (Delta H-0) and entropy (Delta S-0) values of -16,514 kJ/mol and 22.65 J/mol center dot K, respectively.

Phenolic Preservative Removal from Commercial Insulin Formulations Reduces Tissue Inflammation while Maintaining Euglycemia

Summary: This study demonstrated that using Zeolite Y as a filter can effectively remove phenolic preservatives from commercial insulin, reducing their cytotoxicity and inflammation.
