The Role of Cytochrome P450s in Insect Toxicology and Resistance

Structural characterization of the homotropic cooperative binding of azamulin to human cytochrome P450 3A5

Summary: Cytochrome P450 3A4 and 3A5 are important catalysts in the metabolism of therapeutic drugs, and Azamulin is used as a selective inhibitor to define their roles in drug development. Unlike 3A4, 3A5 exhibits homotropic cooperativity in inhibition. The crystal structure of 3A5 with two Azamulin molecules reveals their stacked orientation in the active site. The binding of distal Azamulin causes induced fit and excludes water from hydrophobic surfaces, which is enhanced by the stacking interaction with proximal Azamulin. These findings contribute to understanding cooperative binding and the structural differences between 3A5 and 3A4.

Overexpression of Multiple Cytochrome P450 Genes Conferring Clothianidin Resistance in Bradysia odoriphaga

Summary: Piperonyl butoxide (PBO) significantly increases the toxicity of clothianidin in the clothianidin-resistant population of Bradysia odoriphaga. The population with resistance has higher P450 enzyme activity and overexpression of four P450 genes. Tissue-specific expression analysis suggests that these genes are highly expressed in the midgut and/or Malpighian tubules. Knockdown of three P450 genes increases the susceptibility of B. odoriphaga to clothianidin.

Phylogenomic and functional characterization of an evolutionary conserved cytochrome P450-based insecticide detoxification mechanism in bees

Summary: The validity of using honeybees as a model for other bee species in pesticide risk assessment has been questioned. Recent research has shown that certain enzymes in honeybees play a critical role in insecticide detoxification, but it is unclear if these enzymes are present in diverse bee species. This study identified functional orthologs of these enzymes in various bee species and found that the ability to metabolize certain insecticides is conserved across major bee families, with some exceptions. These findings highlight the potential of using toxicogenomic approaches to assess pesticide risk for nonmanaged bee species.

This study identified the NR2E nuclear receptor HR83 as critical for chlorpyrifos resistance in brown planthoppers, regulating the expression of detoxification genes.
