The bacterium stimulates local upregulation of exocytosis in bacterial protrusions, recruiting exocyst components and regulators to facilitate efficient protrusion formation and cell-to-cell spread. Depletion of exocyst proteins or RalA leads to reduced protrusion formation frequency and length, highlighting the importance of exocyst in controlling both initiation and elongation of protrusions by Shigella.
