Virus interactions with the actin cytoskeleton-what we know and do not know about SARS-CoV-2

Malgorzata Kloc et al.

Summary: The interaction of viruses with the actin cytoskeleton plays an important role in their entry and propagation in host cells. However, there is limited knowledge about the interactions of SARS-CoV-2 with the actin cytoskeleton. Further research in this area could lead to the development of novel antiviral therapies for mitigating the severity of COVID-19.

Mechanisms of SARS-CoV-2 entry into cells

Cody B. Jackson et al.

Summary: The entry of SARS-CoV-2 into host cells is facilitated by the interaction between the viral spike protein and ACE2, leading to virus-cell membrane fusion, which has been extensively studied at the structural and cellular levels worldwide. This understanding has paved the way for the development of effective vaccines in response to the COVID-19 pandemic, caused by the infection with SARS-CoV-2.

Recovery of anosmia in hamsters infected with SARS-CoV-2 is correlated with repair of the olfactory epithelium

Rachel A. Reyna et al.

Summary: This study developed a behavioral test of anosmia in SARS-CoV-2-infected hamsters, and found a moderately strong correlation between the level of anosmia and the score of histological damage within the olfactory epithelium. It also found a moderately strong correlation between the level of anosmia and the thickness of the olfactory epithelium, previously demonstrated to be severely damaged upon infection. This food-searching behavioral test can therefore serve as a simple and effective screening method for various therapeutics for SARS-CoV-2-related anosmia in hamster models.

An intra-cytoplasmic route for SARS-CoV-2 transmission unveiled by Helium-ion microscopy

Antonio Merolli et al.

Summary: This study used scanning Helium-ion microscopy to investigate the transmission mechanism of SARS-CoV-2 between infected cells. The findings revealed a novel intra-cytoplasmic connection, including the presence of nanotubes, cell fusion events, and extracellular vesicles. These findings provide insights into the alternative route of viral transmission and its potential role in evading immune surveillance.

Pyroptosis and Its Role in SARS-CoV-2 Infection

Zsofia Agnes Bittner et al.

Summary: This article mainly introduces the discovery of the pore-forming inflammatory cell death pathway, pyroptosis, and its role in health and disease. The molecular events related to GSDMD-mediated pore formation are emphasized, and the ways of SARS-CoV-2 induced NLRP3 inflammasome formation leading to pyroptosis, which is strongly associated with COVID-19 pathology, are summarized. The potential of NLRP3 and GSDMD inhibitors as therapeutics to counter excessive inflammation is also explored.

The structural basis of accelerated host cell entry by SARS-CoV-2†

Murat Seyran et al.

Summary: SARS-CoV-2 utilizes its S protein to rapidly enter cells, sliding quickly with the flat sialic acid-binding domain in the N-terminal domain interacting with the epithelial layer, and entering via the ACE-2 receptor. The binding between viral RBD and ACE-2 may be optimal, and protein enzymes can accelerate cell entry by SARS-CoV-2.

Determination of the Concentration of IgG against the Spike Receptor-Binding Domain That Predicts the Viral Neutralizing Activity of Convalescent Plasma and Serum against SARS-CoV-2

Llipsy Santiago et al.

Summary: Passive immunization with hyperimmune plasma from convalescent patients has been proposed as a potential treatment for COVID-19, although its efficacy remains uncertain. A standardized method to predict the viral neutralizing activity of plasma against SARS-CoV-2 is crucial. Through an in-house quantitative ELISA test, it was found that most convalescent donors had IgG antibodies specific to the spike receptor-binding domain of SARS-CoV-2, with concentrations correlating with disease severity and gender. This suggests that the concentration of anti-RBD IgG in plasma may be a predictor of its efficacy in neutralizing the virus in vitro.

Tunneling nanotubes: Reshaping connectivity

Chiara Zurzolo

Summary: Tunneling nanotubes (TNTs) are open membranous channels between cells that allow for the diffusion of various cellular materials. While their discovery has enriched the field of cellular communication research, there is confusion surrounding their structure-function and their ability to transfer material between different cell populations. The use of low-resolution imaging methods in most studies has contributed to this inconsistency, and further research is needed to fully understand the properties of TNTs.

Summary: Tunneling nanotubes (TNTs) are filamentous structures that mediate efficient cell-to-cell communication but can also facilitate the spread of various pathogens, including viruses. Research has shown the diverse roles of TNTs in diseases, such as viral infections, neurodegenerative diseases, chemotherapeutic resistance, and cancer pathogenesis.

Analysis of non-structural proteins, NSPs of SARS-CoV-2 as targets for computational drug designing

Resal Raj

Summary: This research utilized computational drug designing to analyze potential drugs targeting NSP proteins of the SARS-CoV-2 genome, identifying three drugs that have promising binding with NSP 3, 5, 11, 14, and 15. The study also found that these three drugs have similar binding pockets with NSPs, indicating a potential for effective targeting of the viral proteins.
