Summary: IP-HILIC-MS with glycine addition in TFA-containing mobile phases enhances MS detection sensitivity for glycopeptides up to 50-fold, by eliminating the ion-suppression effect of the ion-pairing agent while retaining excellent separation capacity. This method confidently identifies more site-specific N-linked glycans for IgG1, IgG4 mAbs, and Fc-domain fusion proteins, while achieving comparable quantitative results compared to traditional methods. Additionally, IP-HILIC-MS can be used for identifying low-level O-glycosylation and non-consensus N-glycosylation on mAbs without enrichment prior to LC-MS analysis.
