Radio Signals from Live Cells: The Coming of Age of In-Cell Solution NMR

Enrico Luchinat et al.

Summary: A detailed understanding of the complex processes that contribute to the functionality of cells and organisms is crucial for disease comprehension and drug development. Among the available structural techniques, solution NMR is the only method capable of investigating macromolecular structures and dynamic behaviors directly within living cells. Over the past 20 years, in-cell NMR has emerged as a powerful tool, allowing researchers to study cell samples, isotopic labeling, and bioreactor devices, thus enhancing the lifetime of cells for real-time monitoring of intracellular metabolites and proteins.

In-cell NMR: From target structure and dynamics to drug screening

Enrico Luchinat et al.

Summary: The cellular environment plays a crucial role in the structure and function of drug targets and their interaction with potential drugs. In-cell NMR spectroscopy allows for direct studies of protein and nucleic acid structures in living cells, providing a unique approach to investigating ligand-target interactions in their native cellular context. When applied to drug screening, in-cell NMR can provide insights into binding kinetics and affinity towards cellular targets, offering a powerful tool for improving drug potency at an early stage of development.

Biogenesis of Iron-Sulfur Clusters and Their Role in DNA Metabolism

Ruifeng Shi et al.

Summary: Iron-sulfur clusters are redox-active protein cofactors that require many components for their synthesis and insertion into target proteins. Mitochondrial ISC assembly cooperates with the cytosolic Fe/S protein assembly system to mature cytosolic and nuclear ISC. ISCs play important roles in DNA metabolism.
