A new experimental approach combining SPASM FRET sensors and GPMV technology was developed to measure and analyze ligand molecular efficacy, confirming the receptor-G-peptide interaction as a significant structural determinant. Leveraging this technology, a two-stage receptor activation mechanism was elucidated through the combination of v-SPASM measurements and molecular dynamics simulations.
