Multiple causal variants underlie genetic associations in humans

Summary: This study evaluates the functional effects of genetic variants in high local linkage disequilibrium for independent cis-expression quantitative trait loci using a massively parallel reporter assay. The results show that 17.7% of the cis-expression quantitative trait loci exhibit multiple major allelic effects in tight linkage disequilibrium. The detected regulatory variants are highly enriched in activating chromatin structures and allelic transcription factor binding. Integration of the reporter assay profiles with colocalizations of cis-expression quantitative trait loci and complex traits across various human traits and diseases reveals sets of causal variants that contribute to genetic association signals.

IFN-gamma and TNF-alpha Gene Polymorphisms in Multiple Sclerosis Patients in Northwest Iran

Summary: The study investigated the allelic and genotypic frequencies of IFN-gamma and TNF-alpha genes in MS patients of Azeri population, finding that the A allele and AT genotype of IFN-gamma +874A/T gene, as well as the G allele and AG genotype of TNF-alpha 308 G/A gene, were more common in MS patients. These gene polymorphisms were considered potential risk factors for MS susceptibility.
