Direct Reprogramming and Induction of Human Dermal Fibroblasts to Differentiate into iPS-Derived Nucleus Pulposus-like Cells in 3D Culture

Shoji Seki et al.

Summary: This study demonstrates the successful induction of nucleus pulposus (NP)-like cells using normal human dermal fibroblasts (NHDFs) through ectopic expression of specific genes in a three-dimensional culture system. These findings may contribute significantly to the development of effective strategies for treating intervertebral disc (IVD) diseases.

Clinical outcomes following intradiscal injections of higher-concentration platelet-rich plasma in patients with chronic lumbar discogenic pain

Cole Lutz et al.

Summary: This study found that intradiscal injections of higher-concentration PRP can significantly improve pain and functional scores in patients with lumbar discogenic pain, and achieve higher satisfaction rates. This suggests that using PRP preparations with a higher concentration of platelets can optimize clinical outcomes.

Correlation between the degree of pain relief following discoblock and short-term surgical disability outcome among patients with suspected discogenic low back pain

Tero Korhonen et al.

Summary: This study evaluated the predictive ability of the degree of pain relief after discoblock on the disability outcome following fusion or TDR surgery. The results showed a correlation between the degree of pain relief and changes in ODI scores, suggesting that discoblock can serve as a useful criterion for assessing disability outcome prior to surgery.

Ketorolac tromethamine represses senescence in aging articular chondrocytes

Wei Zhang et al.

Summary: The study investigated the potential of ketorolac tromethamine in treating osteoarthritis by examining its effects on cellular senescence in chondrocytes. It was found that ketorolac tromethamine repressed cellular senescence by inhibiting COX-2.

Rutin maintains redox balance to relieve oxidative stress induced by TBHP in nucleus pulposus cells

Jian Zhou et al.

Summary: Rutin is found to have a protective effect against oxidative stress in nucleus pulposus cells (NPCs). It can reverse oxidative stress and maintain cellular function of NPCs, suggesting it as a potential therapeutic option for intervertebral disc degeneration.

In vitro and in vivo Effects of a Single Dose of Bupivacaine 5% on Donkey Chondrocytes

Kamal Hussein et al.

Summary: The study found that 5% bupivacaine did not significantly affect the viability of donkey chondrocytes at various time points, and there were no significant differences in biochemical analytes of serum and synovial fluid following IA bupivacaine injection compared with saline injection. Additionally, in vivo IA injection of bupivacaine showed no significant differences in radiography, CT scan, gene expression of cartilage catabolic biomarkers, and histopathological examination, providing evidence for the safety of bupivacaine on donkey cartilage.
