pH-Sensitive Nanoparticles Composed Solely of Membrane-Disruptive Macromolecules for Treating Pancreatic Cancer

Feng Fan et al.

Summary: A new approach using long-circulating, pH-sensitive nanoparticles composed solely of cellular membrane-disruptive molecules has been proposed for treating pancreatic cancer. These nanoparticles effectively permeabilized the stromal barrier and eradicated pancreatic cancer cells without promoting tumor metastasis.

Enhanced Stability and Bioactivity of Natural Anticancer Topoisomerase I Inhibitors through Cyclodextrin Complexation

Victor Gonzalez-Ruiz et al.

Summary: The use of cyclodextrins as drug nano-carrier systems for drug delivery is gaining importance in the pharmaceutical industry due to the interesting pharmacokinetic properties of the resulting inclusion complexes. This study showed that the complexes of camptothecin and luotonin A with cyclodextrins can enhance antitumor activity, suggesting potential for stable analogues of camptothecin in cancer treatment.

Conformable hierarchically engineered polymeric micromeshes enabling combinatorial therapies in brain tumours

Daniele Di Mascolo et al.

Summary: Surgical resection is the primary treatment for glioblastoma multiforme, but the infiltrating nature of the tumor, heterogeneity, and blood-brain barrier hinder drug delivery, leading to recurrence and poor prognosis. A mechanically flexible mesh has been engineered to release nanomedicines and small molecules for tumor therapy, offering a potential solution to these challenges.

The evolution of commercial drug delivery technologies

Ava M. Vargason et al.

Summary: This review article discusses how challenges in delivering small molecules, nucleic acids, peptides, proteins, and cells have led to the development of commercial products and are now guiding the delivery of live-cell therapeutics. Drug delivery technologies have enabled the development of pharmaceutical products that enhance therapeutic delivery, minimize off-target effects, and improve patient compliance. Three drug delivery paradigms have been developed to aid in the successful delivery of small molecules and macromolecules, contributing to the initial clinical success of each therapeutic class.
