Oscillating the local milieu of polymersome interiors via single input-regulated bilayer crosslinking and permeability tuning

Guhuan Liu et al.

Summary: The permselectivity of cellular membranes is crucial for maintaining intracellular homeostasis. However, it is challenging to synergize the stability and permeability of artificial cells and imitate local milieu fluctuations. In this study, a concurrent cross-linking and permeabilizing strategy was developed using pH-responsive polymersomes containing Schiff base moieties within bilayer membranes. The strategy allowed precise control of mesh sizes of the crosslinked bilayers, resulting in discriminative permeability to molecules of specific sizes. The permselectivity of the bilayer membranes enabled intravesicular pH oscillation and the dissipative self-assembly of pH-sensitive dipeptides. Additionally, the permeabilization of polymersomes could be regulated by intracellular pH gradient, allowing controlled release of encapsulated payloads.

Summary: In this study, thermo-responsive and pH-responsive triblock copolymers were synthesized, which can self-assemble into nanostructures and encapsulate hydrophobic molecules under specific conditions. The P4VP blocks can coordinate with Cu2+ to form a thermo- and pH-responsive nanogel, achieving size reduction through thermo- or pH-induced deswelling.

Reversibly self-assembled pH-responsive PEG-p(CL-g-TMC) polymersomes

Pascal L. W. Welzen et al.

Summary: Polymersomes are being explored as drug delivery systems in the biomedical field due to their ability to protect and transport cargo in the body. Incorporating pH-sensitive groups allows for controlled release of cargo at target sites, such as tumor tissues. By modifying the polymer backbone with imidazole moieties, these polymeric vesicles disassemble at mild acidic conditions and reassemble efficiently at higher pH, leading to enhanced encapsulation of cargo. The potential of this polymeric system for future biomedical applications, such as adjuvant delivery, has been demonstrated.
