3,4-Difluorobenzocurcumin Inhibits Vegfc-Vegfr3-Erk Signalling to Block Developmental Lymphangiogenesis in Zebrafish

Kazuhide S. Okuda et al.

Summary: Lymphangiogenesis plays critical roles in disease, and studies have shown that CDF can effectively inhibit lymphangiogenesis by interfering with the VEGFC-VEGFR3-ERK signaling pathway. These findings suggest that CDF may be a candidate for treating metastasis and diseases of aberrant lymphangiogenesis.

Ileitis-associated tertiary lymphoid organs arise at lymphatic valves and impede mesenteric lymph flow in response to tumor necrosis factor

Rafael S. Czepielewski et al.

Summary: The formation of tertiary lymphoid organs (TLOs) in the mesentery is a feature of Crohn's disease, blocking cellular and molecular outflow from the gut and causing lymph leakage and backflow. TNF neutralization at early stages can restore lymph transport, but developed chronic TLOs resist regression and restoration of flow after TNF neutralization. TNF stimulation of lymphatic endothelial cells disrupts immune cell transport and valve-associated gene induction, contributing to Crohn's disease pathology.

Targeted delivery of mycophenolic acid to the mesenteric lymph node using a triglyceride mimetic prodrug approach enhances gut-specific immunomodulation in mice

Ruby Kochappan et al.

Summary: The mesenteric lymph nodes play a crucial role in generating adaptive immune responses and contribute to the pathophysiology of various conditions; targeting immunomodulating drugs to the MLN may be beneficial. Using a triglyceride mimetic prodrug approach, this study found that delivering mycophenolic acid in prodrug form resulted in increased lymphatic transport and MLN concentrations, demonstrating potential pharmacodynamic benefits of targeting the MLN with this technology.

Regulation of Intestinal Inflammation by Dietary Fats

Abigail R. Basson et al.

Summary: With the rise of human obesity, dietary fats have become the focus of biomedical research. High-fat diets, particularly rich in long-chain saturated fatty acids, have pro-inflammatory effects on multiple organs, while certain saturated fats like lauric and myristic acids may have anti-inflammatory and protective properties. Although some effects observed in epidemiological studies may oversimplify the mechanisms, it is essential to understand the complex interactions between the host, specific fatty acids, and genetics/variations of gut microbiota.

Characterization of cytoplasmic lipid droplets in each region of the small intestine of lean and diet-induced obese mice in response to dietary fat

Alyssa S. Zembroski et al.

Summary: This study reveals the dynamic of fat absorption in the small intestine of lean and obese mice in response to dietary fat. Unique features of cytoplasmic lipid droplets (CLDs) were identified in the proximal, middle, and distal regions of the small intestine of lean and obese mice, potentially contributing to regional differences in dietary fat processing, absorption, or CLD metabolism.

Adipocyte inflammation and pathogenesis of viral pneumonias: an overlooked contribution

Pablo C. Alarcon et al.

Summary: Obesity is established as an independent risk factor for increased susceptibility and severity to viral respiratory pneumonias associated with H1N1 influenza and SARS-CoV-2 pandemics. Alterations in immune cell metabolism and function are key factors of dysregulated inflammation, but the contribution of altered adipocytes in obesity to infectious disease pathogenesis is often overlooked.

Coordinating Effect of VEGFC and Oleic Acid Participates to Tumor Lymphangiogenesis

Florent Morfoisse et al.

Summary: This study found that lymphangiogenesis occurs in tumor lesions and surrounding adipose tissue, and the density of lymphatic vessels is correlated with an increase in circulating free fatty acids in the lymph of tumor-bearing mice. Adipocyte-released FFA can stimulate the sprouting of lymphatic endothelial cells, with monounsaturated oleic acid playing a key role in the promotion of lymphangiogenesis in the tumor microenvironment.
