Synthesis, in vitro Antileishmanial Efficacy and Hit/Lead Identification of Nitrofurantoin-Triazole Hybrids

Summary: Leishmaniasis is a common parasitic infection, especially prevalent among populations in low-income developing countries. The lack of effective vaccines means that chemotherapy is the primary treatment method. However, current antileishmanial drugs are limited due to their toxicity and drug-resistant strains. Therefore, there is a need for new and effective antileishmanial agents. In this study, a series of nitrofurantoin-triazole hybrids were synthesized and evaluated for their activity and cytotoxicity. One derivative, featuring a propargyl moiety, showed the highest activity against the causative pathogens of visceral leishmaniasis, with no toxicity to human cells. This compound has potential as an alternative therapy and warrants further investigation.

Mutations in an Aquaglyceroporin as a Proven Marker of Antimony Clinical Resistance in the Parasite Leishmania donovani

Summary: This study identified gene mutations in AQP1 associated with antimony resistance in Leishmania donovani by sequencing the genomes of clinical isolates and conducting experimental validation through CRISPR-Cas9-mediated genome editing. Restoring the wild-type AQP1 in resistant isolates sensitized them to antimonials, while introducing the mutation in sensitive isolates led to antimony resistance. Through genomic analysis and gene editing, the role of AQP1 mutations in clinical resistance was confirmed.

Summary: 5-nitro-furan nitrones (1) and 5-nitro-thiophene nitrones (2) were synthesized in one step and showed potent leishmanicidal activity, particularly against intracellular amastigote forms of Leishmania amazonensis and L. infantum. The redox potential of the compounds was modulated by the nitrone group, which could potentially slow the development of resistance. Oral administration of 1b in mice infected by L. infantum significantly reduced parasite load in the spleen and liver.
