Current Knowledge on Microviridin from Cyanobacteria

Samuel Cavalcante do Amaral et al.

Summary: Microviridins, a class of ribosomal peptides derived from cyanobacteria, are potent protease inhibitors with potential applications in drug development and mosquito control. They also play a key ecological role in defending cyanobacteria against microcrustaceans. This review aims to systematically identify the key characteristics of microviridins, including their chemical structure, biosynthesis, as well as their biotechnological and ecological significance.

Structure Revision of Protoaculeine B, a Post-translationally Modified N-Terminal Residue in the Peptide Toxin Aculeine B

Raku Irie et al.

Summary: The structure of protoaculeine B, a marine peptide toxin, has been revised to a cis-1,3-disubstituted tetrahydro-beta-carboline framework. Two truncated model compounds were prepared using the one-step Pictet-Spengler reaction of tryptophan, which successfully reproduced the natural product's profiles and confirmed the appropriateness of the structure revision.

Leveraging orthogonal mass spectrometry based strategies for comprehensive sequencing and characterization of ribosomal antimicrobial peptide natural products

Tessa B. Moyer et al.

Summary: This article reviews orthogonal approaches to accelerate comprehensive and accurate molecular characterization of ribosomal antimicrobial peptide natural products without the need for prior isolation. Strategies such as chemical derivatization, proteolysis, multistage MS fragmentation, and separation can provide complementary amino acid composition and post-translational modification data to constrain sequence solutions. Case studies of gomesin and styelin D demonstrate the practical implementation of these approaches.
