Obesity risk is associated with altered cerebral glucose metabolism and decreased μ-opioid and CB1 receptor availability

Tatu Kantonen et al.

Summary: This study investigated the association between brain insulin signaling, mu-opioid receptors (MORs), and cannabinoid CB1 receptors with the risk of developing obesity. The results showed that high obesity risk was associated with globally increased insulin-stimulated brain glucose uptake, and familial obesity risk factors were related to decreased availability of MORs and CB1Rs. This suggests that hereditary mechanisms promoting obesity may be mediated partly via insulin, opioid, and endocannabinoid messaging systems in the brain.

Ethnic and sex differences in hepatic lipid content and related cardiometabolic parameters in lean individuals

Kitt Falk Petersen et al.

Summary: The study found that the 95th percentile of hepatic triglyceride (HTG) in lean non-Al individuals was 1.85% and HTG concentrations above this threshold were associated with insulin resistance (IR) and cardiovascular risk factors. Premenopausal women were protected from these changes whereas young, lean Asian Indian (AI) men and women manifested increased HTG content and associated cardiometabolic risk factors.

Insulin Resistance Is Associated With Enhanced Brain Glucose Uptake During Euglycemic Hyperinsulinemia: A Large-Scale PET Cohort

Eleni Rebelos et al.

Summary: In this study, it was found that insulin sensitivity was negatively associated with brain glucose uptake (BGU) during euglycemic hyperinsulinemia, indicating increased BGU in insulin-resistant individuals. Age was negatively related to BGU, while steady-state insulin levels, C-reactive protein and free fatty acid levels, sex, and HbA(1c) were not associated with BGU.

Brain Glucose Metabolism in Health, Obesity, and Cognitive Decline-Does Insulin Have Anything to Do with It? A Narrative Review

Eleni Rebelos et al.

Summary: Using [F-18]-FDG-PET to image brain glucose metabolism has been a valuable tool in diagnosing cognitive disorders like Alzheimer's disease, with recent studies revealing connections between obesity, diabetes, and alterations in brain glucose metabolism. Understanding whether changes in brain glucose metabolism are a key determinant in the pathophysiology of metabolic and neurologic disorders is crucial for advancing research in this field.

Tissue-specific glucose partitioning and fat content in prediabetes and type 2 diabetes: whole-body PET/MRI during hyperinsulinemia

Jan W. Eriksson et al.

Summary: The study utilized a novel imaging approach to quantify tissue-specific glucose metabolism. During the development of T2D, insulin-stimulated glucose disposal was impaired and shifted towards the brain. Changes in glucose handling in the brain and liver fat accumulation may worsen insulin resistance in multiple organs.
