Assessment of Thyroid Endocrine Disruption Effects of Parabens Using In Vivo, In Vitro, and In Silico Approaches

Jiefeng Liang et al.

Summary: This study investigated the developmental toxicity of parabens on zebrafish embryos and larvae, with butyl paraben showing the highest toxicity. Paraben exposure led to mortality, decreased hatching rate, reduced body length, lowered heart rate, and malformation. The study also found that paraben exposure disrupted thyroid hormone levels and gene expression in the hypothalamic-pituitary-thyroid axis, indicating their potential thyroid endocrine disruption effects.

Triadimefon in aquatic environments: occurrence, fate, toxicity, and ecological risk

Lin Hou et al.

Summary: Triadimefon is a widely used triazole fungicide that can be detected in surface waters with high rates of detection. It can cause various adverse effects on organisms, including those related to the thyroid, reproductive system, liver, nervous system, as well as carcinogenicity and teratogenicity. The risk assessment based on joint probability curves indicates that the risk of triadimefon in aquatic environments is low.

Endocrine disruption by azole fungicides in fish: A review of the evidence

Tao Huang et al.

Summary: Azole fungicides have been shown to disrupt the endocrine system in fish, particularly affecting the HPG, HPT, and HPA axis. Some azoles have well-documented endocrine disruptor effects, while others lack data, revealing a knowledge gap in understanding azole toxicity. Further studies are needed to focus on non-steroid endocrine pathways, mechanisms of neuroendocrine disruption, and transgenerational effects of azoles on fish.

Toxic effects of triclocarban on larval zebrafish: A focus on visual dysfunction

Xiao-Fan Chen et al.

Summary: The study found that TCC negatively impacts the visual system of zebrafish larvae, resulting in decreased eye size, reduced retinal layer thickness, and lower ganglion cell density. These effects could potentially disrupt the survival and ecological balance of animals.

Chronic Exposure to Climbazole Induces Oxidative Stress and Sex Hormone Imbalance in the Testes of Male Zebrafish

Xiao-Liang Liao et al.

Summary: Climbazole has potential toxicity on the male reproductive system of zebrafish, inducing testicular cell apoptosis and decreasing sperm production. This study fills the data gap on the reproductive toxicity of climbazole to male zebrafish, and highlights the ecotoxicological application of untargeted metabolomics in biomarker discovery.

Development of a Cell-Based Luciferase Complementation Assay for Identification of SARS-CoV-2 3CLpro Inhibitors

Jonathan M. O. Rawson et al.

Summary: A novel cell-based luciferase complementation reporter assay has been developed to identify inhibitors of SARS-CoV-2 3CL(pro) in a BSL-2 setting. This assay can easily distinguish true 3CL(pro) inhibition from cytotoxicity, reducing false positives during screening and facilitating the identification of more potent inhibitors.

Tipiracil binds to uridine site and inhibits Nsp15 endoribonuclease NendoU from SARS-CoV-2

Youngchang Kim et al.

Summary: SARS-CoV-2 Nsp15 is a uridine-specific endoribonuclease that interferes with the innate immune response and is a potential target for therapeutic intervention. Studies have shown that Tipiracil inhibits the activity of Nsp15 by interacting with the uridine binding pocket. This discovery provides new insights for the development of drugs based on uracil scaffold.

Metabolomics reveals the reproductive abnormality in female zebrafish exposed to environmentally relevant levels of climbazole

Ting Zou et al.

Summary: This study found that exposure to climbazole in zebrafish significantly affects reproductive function, leading to decreased fecundity and inhibited cell development in the ovaries. Metabolomics analysis and other biochemical assays revealed that climbazole may trigger reproductive toxicity through alterations in steroidogenesis and oxidative stress, shedding new light on the mechanism of reproductive dysfunction caused by climbazole.
