Oxidative stress, metallomics and blood toxicity after subacute low-level lead exposure in Wistar rats: Benchmark dose analyses

Dragana Javorac et al.

Summary: Exposure to low doses of lead may affect hematological parameters and lipid profiles, as well as antioxidant enzymes, reactive oxygen species, and metal element levels in the body. Results from the study suggest that copper blood levels and superoxide dismutase activity are particularly sensitive indicators of lead toxicity at low levels of exposure.

A methodology for developing key events to advance nanomaterial-relevant adverse outcome pathways to inform risk assessment

Sabina Halappanavar et al.

Summary: Significant progress has been made in the development of Adverse Outcome Pathways (AOPs) focusing on the toxicity mechanisms of chemicals, but there is a lack of consideration for the size-associated properties of manufactured nanomaterials (MNs) in current AOPs. By utilizing existing nanotoxicology literature, this study established a systematic methodology for identifying key events (KEs) relevant to MNs and addressed the limitations and opportunities in the field. Recommendations were provided for future reporting of nanotoxicology results to expedite the development of AOPs for MNs and aid in regulatory decision making.

The Role of Behavioral Ecotoxicology in Environmental Protection

Alex T. Ford et al.

Summary: Chemicals have long been acknowledged to impact human and wildlife behavior. Recent technological advancements have highlighted the adverse effects of contaminants on organismal behavior and ecological outcomes. Concerns about the lack of consideration of behavior in regulatory ecotoxicology have led to the exclusion of many studies from chemical risk assessments. A workshop with international representatives has resulted in consensus perspectives and recommendations to improve the integration of basic and translational sciences with regulatory practices.

Summary: The study developed a subcellular two-compartment toxicokinetic-toxicodynamic model for zebrafish larvae exposed to four metals, showing that copper had the highest toxicity. The results suggest that the observed variation in toxicity of the four metals was mainly determined by toxicodynamic processes.

Ferroptosis: mechanisms and links with diseases

Hong-fa Yan et al.

Summary: Ferroptosis is a form of cell death that is iron-dependent and involves accumulation of lethal lipid species from lipid peroxidation. This review discusses the regulatory mechanisms of ferroptosis and its connections with iron, lipid, and cysteine metabolism pathways. Additionally, it explores the role of ferroptosis in diseases such as cancer, ischemia/reperfusion, Alzheimer's disease, and Parkinson's disease, and evaluates the potential therapeutic applications of ferroptosis inhibitors in clinical settings.
