Alz-Disc: A Tool to Discriminate Disease-causing and Neutral Mutations in Alzheimer's Disease

Summary: This study constructed a comprehensive dataset of Alzheimer's disease-causing and neutral mutations, and developed a machine learning algorithm based on Bayes network to discriminate between these two types of mutations using sequence information alone. The developed method achieved an accuracy of 89% for independent test set, which is 13% higher than available generic methods. This method is freely available as a web server at https://web.iitm.ac.in/bioinfo2/alzdisc/.

Artificial intelligence framework identifies candidate targets for drug repurposing in Alzheimer's disease

Summary: This study develops a network-based artificial intelligence framework that integrates multi-omics data with protein-protein interactome networks to accurately infer drug targets affected by GWAS-identified variants, leading to the discovery of new therapeutics. Through applying this approach to AD, 103 validated ARGs are identified, and three drugs are found to be significantly associated with decreased risk of AD.

Summary: TFEB as a master regulator of the autophagy-lysosomal pathway holds therapeutic potential for neurodegenerative disorders, with small molecules identified as TFEB activators showing promise in protecting against these diseases. Research is ongoing and the development of specific and potent TFEB activators with ideal brain bioavailability could provide a method for treating neurodegenerative disorders.

Summary: Pharmacological inhibition of phosphodiesterase 4D (PDE4D) shows potential in restoring memory function in Alzheimer's disease (AD), but may lead to adverse effects. Increased expression of specific PDE4D isoforms in AD is associated with changes in DNA methylation and severity of pathology and cognitive impairment. Further studies should investigate the functional roles of specific PDE4D isoforms and the safety and efficacy of selective inhibition for memory restoration in AD.

Anti-LINGO-1 antibody ameliorates cognitive impairment, promotes adult hippocampal neurogenesis, and increases the abundance of CB1R-rich CCK-GABAergic interneurons in AD mice

Summary: This study administered an anti-LINGO-1 antibody to AD mice and found significant improvements in cognitive abilities, adult hippocampal neurogenesis, decreased Aβ deposition, increased neuron numbers (especially GABAergic and CCK-GABAergic interneurons), and reduced GABAergic interneurons expressing LINGO-1 and CB1R in the hippocampus. The results suggest that LINGO-1 plays a crucial role in hippocampal neuron loss in AD, and antagonizing LINGO-1 can effectively prevent this loss.
