GPR43 activation-mediated lipotoxicity contributes to podocyte injury in diabetic nephropathy by modulating the ERK/EGR1 pathway

Jian Lu et al.

Summary: The activation of GPR43 contributes to podocyte injury in diabetic nephropathy by mediating lipotoxicity and modulating the ERK/EGR1 pathway. The study also found that GPR43 gene knockout inhibited cholesterol accumulation and autophagy inhibition. These findings suggest that GPR43 could be a potential target for treating podocyte injury in diabetic nephropathy.

The dietary inflammatory index, obesity, type 2 diabetes, and cardiovascular risk factors and diseases

Rohit Hariharan et al.

Summary: Research suggests that an unhealthy diet is associated with an increased risk of obesity, T2DM, and CVD, but there is limited research on the association between dietary inflammation and T2DM and its risk factors, as well as a lack of interventional studies designed to change the inflammatory potential of diets and study its effect on cardiometabolic risk factors and diseases. Interventions studies are recommended to assess whether changes in DII can independently modulate cardiometabolic risk factors and diseases.

The Updates of Podocyte Lipid Metabolism in Proteinuric Kidney Disease

Yu Sun et al.

Summary: Disorder of podocyte lipid metabolism is closely associated with various proteinuric kidney diseases, excessive lipid accumulation can lead to cellular dysfunction, ultimately resulting in cell hypertrophy, detachment, and death. Targeting podocyte lipid metabolism may represent a novel therapeutic strategy for patients with proteinuric kidney disease.

Rotten to the Cortex: Ceramide-Mediated Lipotoxicity in Diabetic Kidney Disease

Rebekah J. Nicholson et al.

Summary: Diabetic kidney disease (DKD) is a prevalent and progressive comorbidity of diabetes mellitus that increases the risk of renal failure. Research suggests that bioactive ceramides may serve as a potential therapeutic target for DKD, as modulation of renal ceramide levels could improve kidney function and ameliorate DKD pathology. Studies have shown that circulating sphingolipid profiles can distinguish DKD patients from diabetic controls, highlighting the potential of ceramide as a central and therapeutically tractable lipid mediator of DKD.

Summary: The kidney relies on specialized epithelial cells in the renal tubules for reabsorption, with the proximal tubules using fatty acid oxidation as the preferred metabolic pathway. After kidney injury, there are alterations in metabolism leading to decreased fatty acid oxidation and increased lactic acid generation.
