Embryo-Endosperm Interactions

Nicolas M. Doll et al.

Summary: Successful seed development and germination require constant interactions between the embryo and endosperm across their shared interface, which affects molecular diffusion and communication. The embryo-endosperm interface is a complex and poorly understood compound apoplast.

Resetting of the 24-nt siRNA landscape in rice zygotes

Chenxin Li et al.

Summary: In plants, zygotes exhibit extensive maternal carryover and a lack of paternal contribution in terms of siRNAs. Zygote formation involves widespread redistribution of 24-nt siRNAs. The transition to totipotency in plant zygotes is accompanied by resetting of epigenetic reprogramming.

The miRNome function transitions from regulating developmental genes to transposable elements during pollen maturation

Cecilia Oliver et al.

Summary: Animal and plant microRNAs (miRNAs) are essential for regulating development. Plant miRNAs not only target genes, but also regulate transposable elements (TEs). However, the role of miRNAs and their target mRNAs during pollen development is still poorly understood.

RNA Pol IV induces antagonistic parent-of-origin effects on Arabidopsis endosperm

Prasad R. V. Satyaki et al.

Summary: This study reveals the important role of plant-specific RNA polymerase IV (Pol IV) in regulating endosperm gene expression. Maternally and paternally acting Pol IV have distinct effects on endosperm, and may be related to the evolution of a gene regulatory system under parental conflict.

DNA methylation dynamics of sperm cell lineage development in tomato

Yunlong Lu et al.

Summary: The study found that the DNA methylation of sperm cells (SCs) in tomato is closely related to the generative cell (GC) and that the methylation landscape in SCs may be pre-established following the asymmetric division or inherited from the GC. The impact of DNA methylation on transcription varies in different cell types, with gene and transposable element (TE) expression in the microspore being more predictable by DNA methylation.

Male fertility in Arabidopsis requires active DNA demethylation of genes that control pollen tube function

Souraya Khouider et al.

Summary: This study reveals the essential role of active DNA demethylation in regulating genes involved in pollen function. Six pollen-specific genes show decreased expression and increased DNA methylation in dme and dme;ros1 mutants. Reinstalling the expression of these genes individually improves male fertility.

Epigenetic reprogramming rewires transcription during the alternation of generations in Arabidonsis

Michael Borg et al.

Summary: The alternation between morphologically distinct haploid and diploid life forms in plants and algae is controlled by epigenetic modifications such as the loss of H3K9me2 and global loss of H3K27me3. These events lead to significant changes in chromatin accessibility and transcriptional reprogramming, guiding the transition between generations in flowering plants.

Independent parental contributions initiate zygote polarization in Arabidopsis thaliana

Kai Wang et al.

Summary: Embryogenesis of flowering plants is initiated by polarization of the zygote, with various protein signaling pathways involved. These pathways include MAP kinase signaling pathway, ESF1 family of cysteine-rich peptides, and the YDA pathway. Furthermore, there is a speculated parental tug-of-war controlling resource allocation toward the embryo.

DNA demethylases remodel DNA methylation in rice gametes and zygote and are required for reproduction

Shaoli Zhou et al.

Summary: The study revealed that DNA methylation is locally reconfigured after fertilization and intensified during embryogenesis in rice. Three rice DNA glycosylases, DNG702, DNG701, and DNG704, were found to demethylate DNA at distinct genomic regions in gametes and zygotes, playing a crucial role in zygotic gene expression and development. The results suggest that active DNA demethylation in gametes and zygotes is critical for gene expression and reproduction in rice.

Embryo CHH hypermethylation is mediated by RdDM and is autonomously directed in Brassica rapa

Tania Chakraborty et al.

Summary: In Brassica rapa, embryos are hypermethylated in both euchromatin and heterochromatin, which is dependent on RdDM. Contrary to current models, hypermethylation in B. rapa embryos is not associated with demethylation of the endosperm. Maternal somatic RdDM does not play a significant role in global embryo hypermethylation, and there is no clear evidence of maternal influence over embryo methylation at any locus.

Transgenerational effect of mutants in the RNA-directed DNA methylation pathway on the triploid block in Arabidopsis

Zhenxing Wang et al.

Summary: Our study found that the ability of mutants in the RdDM pathway to suppress the triploid block depends on their degree of inbreeding. While first homozygous generation mutants in RdDM components NRPD1, RDR2, NRPE1, and DRM2 have weak or no ability to rescue the triploid block, they are able to suppress the triploid block with successive generations of inbreeding. Inbreeding of nrpd1 was connected with a transgenerational loss of non-CG DNA methylation on sites jointly regulated by CHROMOMETHYLASES 2 and 3.

Transcriptional and imprinting complexity in Arabidopsis seeds at single-nucleus resolution

Colette L. Picard et al.

Summary: Seeds are crucial for plants' life cycle and agriculture, serving as a primary source of calories for humans. Using single-nucleus RNA sequencing, a transcriptional atlas of Arabidopsis thaliana seeds was generated, focusing on the endosperm. Transcriptionally uncharacterized nuclei types were identified in the chalazal endosperm, suggesting spatial and temporal heterogeneity of imprinting. Parental conflict, proposed to drive imprinting evolution, appears strongest at the boundary between filial and maternal tissues in the chalazal region.
