Reverse transcription recombinase polymerase amplification assay for rapid detection of the cucurbit chlorotic yellows virus

Lianyi Zang et al.

Summary: In this study, a new method for rapid detection of Cucurbit chlorotic yellows virus (CCYV) was developed using RPA technology. The analysis could be performed under constant temperature conditions in just 20 minutes without the need for a thermal cycler. The application of this method in field-collected melon samples showed consistent results with the traditional RT-PCR detection.

Summary: In this study, a real-time fluorescent RT-RAA method was developed for rapid and specific detection of ALV-J in poultry. The method showed high sensitivity, good repeatability, and could complete the test within a short time. Moreover, there was no cross-reactivity with other avian leukosis virus subgroups. The study results indicated that this method is simple, easy to use, and suitable for clinical and laboratory diagnosis, providing technical support for the prevention and control of ALV-J.

Rapid and sensitive recombinase polymerase amplification combined with lateral flow strips for detecting Candida albicans

Fang Wang et al.

Summary: Due to modern lifestyles and increasing medical intervention, clinical infections caused by conditionally pathogenic fungi, especially Candida albicans, are becoming more serious. Recombinase polymerase amplification (RPA) combined with lateral flow strips (LFS) is a promising method for rapid and accurate detection of this pathogenic fungus. However, the simplicity of the method brings the risk of false-positive signals from primer-dimers. This study successfully eliminated primer-dependent artifacts in the RPA reaction by using probes, introducing specific base substitutions, and analyzing primer-probe complexes, leading to an improved RPA-LFS system for specific detection of C. albicans within 25 minutes without interference.
