Ssu72 deficiency leads to hyperresponsiveness in T cells upon TCR stimulation due to increased ZAP-70 phosphorylation, which can be restored by reducing ZAP-70 phosphorylation levels. Additionally, Cd4-CreSsu72fl/fl mice show defects in thymic development and alterations in peripheral T cell populations, with a predisposition for spontaneous inflammation.

Expansion of circulating peripheral TIGIT+CD226+CD4 T cells with enhanced effector functions in dermatomyositis

Summary: The T cell Ig and ITIM domain (TIGIT)/CD226 pathway plays a critical role in regulating T cell responses and has shown promise as an immunotherapeutic target in cancer. In this study, we identified and characterized four distinct T cell subsets based on TIGIT/CD226 co-expression in patients with dermatomyositis (DM). The elevated frequency of TIGIT+CD226+ CD4 T cells with enhanced effector function in DM patients, especially those with interstitial lung disease, suggests a potential therapeutic target in blocking CD226 to suppress this subset.
