Copper induces cell death by targeting lipoylated TCA cycle proteins

Peter Tsvetkov et al.

Summary: Copper is an essential cofactor for all organisms, but excessive copper can lead to cell death. We found that copper-dependent cell death mechanism is different from known mechanisms, and it is related to mitochondrial respiration. Copper directly binds to lipoylated components of the tricarboxylic acid cycle, causing protein aggregation and loss of iron-sulfur cluster proteins, ultimately leading to cell death.

Pyroptosis-related lncRNAs are potential biomarkers for predicting prognoses and immune responses in patients with UCEC

Jinhui Liu et al.

Summary: Two clusters were identified, with one showing better prognosis and higher infiltration level; A prognostic signature was established through multiple Cox regression analysis, accurately predicting UCEC; High risk score was associated with reduced immune infiltration and T cells exhaustion, while patients with low risk score had a sensitive response to immunotherapy and chemotherapy.

Connecting copper and cancer: from transition metal signalling to metalloplasia

Eva J. Ge et al.

Summary: Copper is a essential nutrient with both beneficial and toxic properties to cells. Recent progress in transition metal signaling has led to new connections between researchers from different disciplines, with potential for translating basic research into clinical therapies for diseases. The relationship between copper and cancer is particularly important, with opportunities for leveraging disease vulnerabilities through copper-dependent signaling pathways.

Cytotoxic CD8+T cells in cancer and cancer immunotherapy

Hans Raskov et al.

Summary: The functions and interactions between the innate and adaptive immune systems play a crucial role in anticancer immunity. Cytotoxic T cells with CD8 as the most effective effectors in the anticancer immune response form the basis of successful cancer immunotherapies. Advancements in genetically modified or synthetic receptors on cytotoxic T cells are being developed for future cancer treatment. Combinatory regimens may optimize treatment effects and reduce adverse events in cancer therapy.

Akt regulates RSK2 to alter phosphorylation level of H2A.X in breast cancer

Zhi-Feng Guo et al.

Summary: This study investigated the relationship between Akt and H2A.X in human breast cancer cells. It found that Akt negatively regulated the phosphorylation of H2A.X, promoting cellular survival and migration. The findings shed light on the pathogenesis and treatment mechanisms of breast cancer.

Genetic and molecular subtype heterogeneity in newly diagnosed early-and advanced-stage endometrial cancer

Arnaud Da Cruz Paula et al.

Summary: This study aimed to characterize and compare the molecular subtypes and profiles of 175 prospectively-accrued newly diagnosed early-and advanced-stage endometrial cancers. The results showed molecular subtypes present across histologic types, potentially helping stratify EC patients for prognostic and therapeutic purposes. Advanced-stage endometrial cancers had higher genomic complexity and certain genetic mutations were found across different histologic types.

IgA transcytosis and antigen recognition govern ovarian cancer immunity

Subir Biswas et al.

Summary: Most ovarian cancers are infiltrated by activated T cells that have prognostic relevance, but they show low response rates to immune checkpoint inhibitors. On the contrary, memory B cell and plasma cell infiltrates associated with better outcomes may exert protective effects against tumor cells through B cell-derived IgA in ovarian cancer.

Summary: This study proposed a novel stemness-based classification of GBM, dividing patients into two distinct subtypes based on their stemness signature, with patients in Stemness Subtype I being more likely to respond to immunotherapy. The research also investigated the genomic variations and tumor immune microenvironment patterns between the two stemness subtypes.

A risk model developed based on tumor microenvironment predicts overall survival and associates with tumor immunity of patients with lung adenocarcinoma

Jie Wu et al.

Summary: This study investigated genes associated with tumor microenvironment in lung adenocarcinoma and developed a TMErisk score for predicting patient survival. Results showed that high TMErisk was associated with poor overall survival and lower abundance of immune and stromal cells, as well as decreased expression of HLA family genes and immune checkpoint genes. Mutations in certain genes were more common in high-TMErisk group, potentially affecting response to immunotherapy. A lower TMErisk score may indicate better response and overall survival outcomes in immunotherapy.
