Present and future of microglial pharmacology

Eva Simoncicova et al.

Summary: Microglia, as brain resident immune cells, play a crucial role in the development, activity, and plasticity of the central nervous system. They have been identified as potential targets for new neurotherapies due to their involvement in various neurological pathologies. Current research focuses on understanding the complex heterogeneity of microglia and developing more targeted treatments. Although the current therapies have limitations, clinical investigations are underway, and the future of microglial therapeutic research looks promising.

Microglia phagocytose oligodendrocyte progenitor cells and synapses during early postnatal development: implications for white versus gray matter maturation

Muhammad Irfan et al.

Summary: Microglia play a crucial role in normal brain development by engulfing oligodendrocyte progenitor cells and regulating axon ensheathment to influence circuit development. Their unique amoeboid morphology undergoes changes over time, engulfing OPCs during a critical period to maintain proper myelin sheath ratios.

Developmental landscape of human forebrain at a single-cell level identifies early waves of oligodendrogenesis

David van Bruggen et al.

Summary: The study identified the emergence of the first wave of oligodendrocyte lineage cells in the human forebrain as early as the 8th week of gestation, indicating evolutionary conservation of this process between mice and humans. Through analyzing the spatial expression of 50 key genes, regions in the human ventral fetal forebrain where oligodendrogenesis first occurs were identified.

Summary: The blood-brain barrier (BBB) is a structural barrier that regulates communication between the brain and blood to maintain homeostasis. In diabetic encephalopathy, BBB impairment leads to leakage and dysfunction of various cell types. However, the mechanisms and consequences of this impairment are not well understood.

M2 microglia-derived extracellular vesicles promote white matter repair and functional recovery via miR-23a-5p after cerebral ischemia in mice

Yongfang Li et al.

Summary: Research suggests that extracellular vesicles (EVs) derived from M2 microglia can promote white matter repair after cerebral ischemia through the direct targeting of Olig3 by miR-23a-5p. This finding provides a novel therapeutic strategy for white matter repair in stroke and demyelinating diseases.

Microglial vesicles improve post-stroke recovery by preventing immune cell senescence and favoring oligodendrogenesis

Stefano Raffaele et al.

Summary: The activation of microglia and monocyte-derived macrophages affects the regenerative process of oligodendrocyte precursor cells (OPCs), promoting functional recovery after stroke. Regenerative microglia-derived extracellular vesicles (EVs) can restore protective immune cell functions, limit senescence, and enhance OPC maturation, potentially leading to improved neurological functionality in post-stroke phase. Tumor necrosis factor carried by EVs mediates pro-differentiating effects on OPCs, suggesting implications for regenerative therapies.

Direct reprogramming of oligodendrocyte precursor cells into GABAergic inhibitory neurons by a single homeodomain transcription factor Dlx2

Linda L. Boshans et al.

Summary: The study demonstrates that by misexpressing Dlx2 alone in postnatal mouse OPCs, their fate can be switched to GABAergic neurons within 2 days by downregulating Olig2 and upregulating a network of inhibitory neuron transcripts. After two weeks, some OPC-derived neurons generated trains of action potentials and formed clusters of GABAergic synaptic proteins.

Regulation of myelination by microglia

Erin N. Santos et al.

Summary: Recent research has shown that microglia play a crucial role in modulating myelination under normal physiological conditions, a role that has often been overlooked. Microglia achieve this by phagocytosis of cells and myelin membrane, as well as releasing trophic factors, cytokines, and chemokines to influence early oligodendrogenesis, myelin formation, and removal of aberrant myelin during development and throughout life.

TNFR2 Signaling Regulates the Immunomodulatory Function of Oligodendrocyte Precursor Cells

Haritha L. Desu et al.

Summary: The study reveals that neuroinflammation in multiple sclerosis leads to a shift in oligodendrocyte precursor cells (OPCs) towards an immunomodulatory phenotype, reducing their remyelination capacity and impairing their repair function. Furthermore, enhancing TNFR2 activation could be an effective strategy to restore OPC reparative capacity.
