Targeted Liquid Biopsy Using Irradiation to Facilitate the Release of Cell-Free DNA from a Spatially Aimed Tumor Tissue

Jae Myoung Noh et al.

Summary: This study investigated the feasibility of using an anatomically localized, target-enriched liquid biopsy (TLB) in mouse models of lung cancer. The results showed that irradiation could enhance the detection sensitivity of specific mutations in circulating tumor DNA (ctDNA) in the plasma samples of xenograft mice. Furthermore, the use of targeted deep sequencing of ctDNA in lung cancer patients undergoing radiotherapy revealed an increase in ctDNA levels after treatment.

Clinical Practice Guidelines for Pre-Analytical Procedures of Plasma Epidermal Growth Factor Receptor Variant Testing

Saeam Shin et al.

Summary: Standardization of pre-analytical procedures is essential for obtaining clinically reliable results in cfDNA testing. This article provides evidence-based clinical practice guidelines for the pre-analytical phase procedures of plasma EGFR variant testing. By standardizing pre-analytical procedures, the analytical performance of cfDNA testing can be improved.

p16 methylation is a potential predictive marker for abemaciclib sensitivity in gastric cancer

Hyun Joo Bae et al.

Summary: The study found that in gastric cancer cells, EBV and MSI-H type cells with p16 methylation were sensitive to abemaciclib, while genomically stable and chromosomal instability type cells with p16 methylation and intact Rb showed responsiveness to the drug. Additionally, patients with p16 methylation in GC often had a poor prognosis.

Pan-cancer circulating tumor DNA detection in over 10,000 Chinese patients

Yongliang Zhang et al.

Summary: The study conducted a ctDNA-based analysis on over 10,000 pan-cancer Chinese patients, revealing age-related CH variants, detection rates of ctDNA in different cancer types, and the frequencies of potential driver genes. The findings may be valuable for identifying therapeutic targets and developing combined treatment strategies.

Evaluating the analytical validity of circulating tumor DNA sequencing assays for precision oncology

Ira W. Deveson et al.

Summary: This study evaluated the performance of five industry-leading ctDNA assays and found that all assays showed high sensitivity, precision, and reproducibility above 0.5% variant allele frequency. However, below this limit, detection became unreliable, especially when input material was limited. The key challenge for ctDNA assays is the reliable sampling of rare ctDNA fragments, with missed mutations being more common than erroneous candidates.
