The value of single-molecule real-time technology in the diagnosis of rare thalassemia variants and analysis of phenotype-genotype correlation

Shiqiang Luo et al.

Summary: Comparing single-molecule real-time technology (SMRT) with conventional genetic diagnostic technology for rare thalassemia mutations, a study was conducted on 434 cases using both methods. SMRT was able to identify a higher number of variants compared to conventional technology, with a 9.91% increase in positive detection rate. The combination of both methods detected 485 positive alleles among 49 types of variant, and SMRT was able to identify some variants not detected by conventional methods, highlighting its significance in clinical thalassemia gene screening.

Comprehensive Analysis of Spinal Muscular Atrophy SMN1 Copy Number, Intragenic Mutation, and 2+0 Carrier Analysis by Third-Generation Sequencing

Shuyuan Li et al.

Summary: Population-wide carrier screening for spinal muscular atrophy (SMA) is recommended, but current methods focus on SMN1 copy number and fail to identify carriers with pathogenic intragenic mutations and silent carriers. Researchers developed a method called CASMA that accurately detects SMA carriers, improves detection rates, and reduces residual risk in SMA carrier screening.

Comprehensive Analysis of Congenital Adrenal Hyperplasia Using Long-Read Sequencing

Yingdi Liu et al.

Summary: This study developed a comprehensive analysis method for congenital adrenal hyperplasia (CACAH) using long-read sequencing, which accurately identified pathogenic variants in CAH candidate genes. Compared to traditional methods, CACAH showed higher sensitivity and specificity, providing a comprehensive approach for CAH genetic diagnosis and carrier screening.

Beyond Trinucleotide Repeat Expansion in Fragile X Syndrome: Rare Coding and Noncoding Variants in FMR1 and Associated Phenotypes

Cedrik Tekendo-Ngongang et al.

Summary: Various variants of the FMR1 gene have been shown to cause disease phenotypes, including splicing variants, missense variants, full deletions, nonsense variants, and frameshift variants. In addition, FMR1 deletions may not only occur in patients with mosaic full mutations, but also in patients with normal-sized CGG repeats, suggesting genomic instability in the CGG repeat region. Clinical tests for potential FMR1-related indications should include methods capable of detecting small coding, noncoding, and copy number variants.

Laboratory testing for fragile X, 2021 revision: a technical standard of the American College of Medical Genetics and Genomics (ACMG)

Elaine Spector et al.

Summary: Molecular genetic testing of the FMR1 gene is commonly performed in clinical laboratories to identify pathogenic variants associated with fragile X syndrome and other related conditions. This document provides updated information on FMR1 pathogenic variants, including prevalence, genotype-phenotype correlations, and variant nomenclature, as well as methodological considerations for testing. The American College of Medical Genetics and Genomics Laboratory Quality Assurance Committee plays a role in maintaining high technical standards for genetic tests, including those related to fragile X syndrome.

Characterization of FMR1 Repeat Expansion and Intragenic Variants by Indirect Sequence Capture

Valentina Grosso et al.

Summary: In this study, Xdrop technology coupled with Nanopore and Illumina sequencing was used to characterize the FMR1 gene related to FXS. By efficiently enriching and analyzing large DNA fragments, the study successfully enabled parallel analysis of repeat expansions and SNVs/indels at single-nucleotide resolution, improving diagnosis efficiency and genetic counseling for FXS patients and their relatives.

Summary: ASD is a behavioral dysfunction affecting about 2% of the US population, with a substantial genetic heterogeneity. FXS is the most common single-gene disorder associated with ASD, caused by the expansion of an unstable CGG repeat in the FMR1 gene on chromosome X.
