Early aggregation mechanism of Aβ16-22 revealed by Markov state models

Summary: By using molecular dynamics simulations and Markov state model, the early oligomerization mechanism of A beta(16-22) peptides was investigated. It was found that the dimeric form of the peptides adopted globular random-coil or extended beta-strand like conformations, and intermolecular antiparallel beta-sheets were the most common type of secondary structure in the observed dimers. The states containing beta secondary structure and extended coiled structures were majorly involved in aggregation.

Segmental structural dynamics in Aβ42 globulomers

Summary: β amyloid 42 plays a central role in the pathogenesis of Alzheimer's disease, forming toxic oligomers in addition to insoluble fibrils. Our study on the structural dynamics of β amyloid 42 globulomers revealed residues 31-34 as the most stable segment, suggesting a lack of well-packed structural core in the globulomers.

Summary: Evidence suggests that the neuronal cell membrane is the main site of oligomer-mediated neuronal toxicity in Alzheimer's disease caused by amyloid-beta peptides. Interactions between amyloid-beta oligomers and the neuronal membrane decrease the propensity of beta-sheet formation in the oligomers. Gangliosides GM1 are identified as the main lipid interaction partners of A beta 42.

Kinetic and thermodynamic stability comparison for the fibrillar form of small amyloid-β(1-42) oligomers using scaled molecular dynamics

Summary: This study investigates the kinetic and thermodynamic stabilities of small A beta oligomers in fibrillar conformations to understand when these aggregates start forming stable fibrils. The research findings suggest that the kinetic stability of oligomers increases with size, particularly beyond five-chain oligomers at the C-terminus end. It was also observed that fully stable fibril formation may require aggregates larger than those considered in the study.

The analyses presented in this study will serve as a useful reference guide for understanding the behavior of Aβ peptide and other similar disordered proteins.
