Molnupiravir and Its Antiviral Activity Against COVID-19

Lili Tian et al.

Summary: Molnupiravir is a broad-spectrum antiviral drug with the potential to treat and prevent various coronaviruses, including SARS-CoV-2. Clinical trials have shown its beneficial effects and favorable safety profile for mild to moderate COVID-19 patients. Molnupiravir has the potential to be a therapeutic candidate against COVID-19.

Inhibitors of dihydroorotate dehydrogenase cooperate with molnupiravir and N4-hydroxycytidine to suppress SARS-CoV-2 replication

Kim M. Stegmann et al.

Summary: This study demonstrates a strong synergy between the nucleoside analog N4-hydroxycytidine (NHC) and inhibitors of dihydroorotate dehydrogenase (DHODH) in inhibiting the replication of SARS-CoV-2. Inhibition of DHODH increases the incorporation of NHC into viral RNA, leading to defective virus genomes. The combination of NHC and DHODH inhibitors may improve therapy options for COVID-19.

A Systematic Study on the Optimal Nucleotide Analogue Concentration and Rate Limiting Nucleotide of the SARS-CoV-2 RNA-Dependent RNA Polymerase

Hasan Vatandaslar

Summary: The current COVID-19 pandemic has emphasized the need for more efficient antiviral compounds. The effectiveness of adenosine-based analogs, commonly used drugs for inhibiting SARS-CoV-2 RNA-dependent RNA polymerase (RdRp), is influenced by endogenous adenosine levels and the approved concentrations for COVID-19 treatment may not be optimal. Additionally, GTP has been identified as the rate-limiting nucleotide for SARS-CoV-2 replication.

Summary: The need for new drug treatments for COVID-19, caused by SARS-CoV-2, is highlighted due to the virus's continued evolution. One potential strategy is the combination of BRQ and DPY to enhance depletion of the cellular pyrimidine nucleotide pool and show antiviral activity against SARS-CoV-2 variants.

Summary: CTPS1 is essential for an effective immune response, with deficiency leading to severe immunodeficiency. CTPS1 expression is up-regulated in activated lymphocytes to meet the increased demand for nucleic acid and lipid synthesis. Selective inhibition of CTPS1 could be beneficial for treating immune disorders and lymphocyte cancers, with recent small-molecule inhibitors showing promise in inhibiting proliferation of human T cells.

Nucleoside Analogs and Nucleoside Precursors as Drugs in the Fight against SARS-CoV-2 and Other Coronaviruses

Nicola Borbone et al.

Summary: Coronaviruses are a family of enveloped RNA viruses that cause a wide range of respiratory infections in mammals, including humans. Three human CoV species have led to severe respiratory diseases such as SARS, MERS, and the current COVID-19 pandemic. The emergence of SARS-CoV-2 in late 2019 has resulted in a global public health crisis, highlighting the urgent need for effective drugs to combat the infection. The repurposing of nucleoside derivatives is a promising strategy for quickly developing therapies against SARS-CoV-2 and other HCoV species.

Molnupiravir promotes SARS-CoV-2 mutagenesis via the RNA template

Calvin J. Gordon et al.

Summary: The study found that Molnupiravir or NHC can increase the mutation frequencies in replicating coronaviruses, leading to enhanced antiviral effects; NHC-TP primarily competes with CTP for incorporation, resulting in RNA mutations; Biochemical data support a mechanism of action of Molnupiravir that is primarily based on RNA mutagenesis mediated via the template strand.

Florian Kabinger et al.

Summary: Molnupiravir is an oral antiviral drug candidate that increases viral RNA mutations frequency and inhibits replication of SARS-CoV-2 by altering the substrate preference of RdRp. This two-step mutagenesis mechanism can explain the broad-spectrum antiviral activity of molnupiravir, making it a promising COVID-19 treatment option.
