Electrochemical/visual microfluidic detection with a covalent organic framework supported platinum nanozyme-based device for early diagnosis of pheochromocytoma

Xiaoya Liu et al.

Summary: In this study, a dual-mode (electrochemical/visual) microfluidic device was developed for the rapid and sensitive detection of circulating tumor cells (CTCs) in pheochromocytoma (PCC). The device utilized specific immunogenic binding and nanozymes for capture, separation, and quantification of PCC-CTCs. The device demonstrated a low detection limit and a wide linear range, making it a portable and reliable platform for point-of-care diagnostics.

Full-active Cu2O/drug core/shell nanoparticles based on grafting from drug coordination polymerization combined with PD-1 blockade for efficient cancer therapy

Guanghui Hou et al.

Summary: In this study, a pioneering grafting from drug coordination polymerization strategy was proposed to develop a fully-coordinated and full-active Cu2O/drug core/shell nanoparticles for multimodal cancer therapy. The nanoplatform exhibited a rapid acid-responsive dissociation behavior and released chemotherapeutic drug cisplatin (CDDP) and photosensitizer indocyanine green (ICG) to achieve efficient triple combined photodynamic therapy/chemodynamic therapy/chemotherapy. The combination of the nanoplatform and alpha PD-1 effectively inhibited distal tumor growth, lung metastasis, and tumor recurrence through reprogramming tumor microenvironment and activating CD8+ T cell antitumor immunity. This study provides a new direction for the design of nanodrug delivery systems.

Nanocatalytic Theranostics with Glutathione Depletion and Enhanced Reactive Oxygen Species Generation for Efficient Cancer Therapy

Lian-Hua Fu et al.

Summary: Chemodynamic therapy (CDT) is a promising method for killing cancer cells by converting intracellular hydrogen peroxide (H2O2) into highly toxic hydroxyl radicals ((OH)-O-center dot). An intelligent nanocatalytic theranostics, PGC-DOX, was developed to address the limitations of endogenous H2O2 and high GSH levels in tumor cells, leading to efficient cancer therapy. The system combines H2O2 self-supply, GSH-elimination, and DOX-induced chemotherapy to effectively inhibit tumor growth with minimal side effects in vivo.

Manganese-Dioxide-Coating-Instructed Plasmonic Modulation of Gold Nanorods for Activatable Duplex-Imaging-Guided NIR-II Photothermal-Chemodynamic Therapy

Ting He et al.

Summary: A plasmonic modulation strategy of gold nanorods through manganese dioxide coating has been developed for NIR-II photoacoustic/magnetic resonance imaging-guided photothermal chemodynamic therapy. The LSPR peak of gold nanorods can be tuned by adjusting the thickness of the MnO2 layer, and the MnO2 layer can degrade in the tumor microenvironment for MR imaging and chemodynamic therapy. This strategy can be applied to other metal nanomaterials for construction of a new class of NIR-II nanotheranostics.

Summary: The GCAH nanotheranostic system is targeted towards cancer cells with overexpressed glycoprotein CD44, utilizing glucose oxidase (GOx) as a biotemplate and modifying it with l-Arginine (L-Arg) and hyaluronic acid (HA). It effectively depletes glucose nutrients in the tumor region through GOx-triggered glucose oxidation and enhances gas therapy by oxidizing L-Arg into NO under the acidic tumor microenvironment. The synergistic therapeutic effect of starvation therapy and gas therapy is significant without toxic side effects in 4T1 tumor-bearing mice.
