Stabilization of Spine Synaptopodin by mGluR1 Is Required for mGluR-LTD

Luisa Speranza et al.

Summary: This study found that mGluR-LTD occurs in spines containing Synaptopodin/SA, while spines lacking Synaptopodin/SA are selectively lost. This process is dependent on mGluR1 and can selectively preserve stronger spines while eliminating weaker ones through mechanisms like physical retention and protein degradation.

Impaired response to sleep deprivation in heterozygous Disc1 mutant mice

Chih-Yu Tsao et al.

Summary: This study found that while the sleep pattern and circadian activity were not altered in Disc1 Het mice, there were differential responses to sleep deprivation (SD) stress between genotypes. Increased microglial density and reduced neuronal proliferative activity were observed in Het-SD mice, suggesting a potential role of genetic factors in mediating responses to SD stress.

Genotype-dependent epigenetic regulation of DLGAP2 in alcohol use and dependence

Weida Meng et al.

Summary: This study identified a specific differentially methylated region, DMR-DLGAP2, associated with alcohol dependence through epigenome-wide association analysis. Methylation within DMR-DLGAP2 was found to be genotype-dependent, allele-specific and associated with reward processing in the brain. The results suggest that DLGAP2 may be a key player in the genetic and epigenetic control of alcohol use and dependence.

Executive functions in youths with autism spectrum disorder and their unaffected siblings

Guan-Jye Seng et al.

Summary: The study found that youths with ASD showed significant deficits in multiple executive function tasks, with potential cognitive endophenotypes in verbal and spatial working memory. Unaffected siblings generally performed normally in executive functions, except for deficits in verbal and spatial working memory.

Learning and reaction times in mouse touchscreen tests are differentially impacted by mutations in genes encoding postsynaptic interacting proteins SYNGAP1, NLGN3, DLGAP1, DLGAP2 and SHANK2

Alexa E. Horner et al.

Summary: The study reveals that mutations in genes encoding key members of the PSD-95 protein complex continue to play a role in neurodevelopmental disorders, affecting cognitive function and behavioral phenotypes. Mouse models suggest that different components of the PSD-95 protein complex have differential regulatory effects on learning, cognitive flexibility, and reaction times.

Shankopathies in the Developing Brain in Autism Spectrum Disorders

Yukti Vyas et al.

Summary: The SHANK family of proteins plays crucial roles at excitatory glutamatergic synapses, with mutations in SHANKs being associated with autism spectrum disorder (ASD). Research shows that lack of SHANK during development can lead to permanent changes in brain wiring, affecting various brain regions and synaptic functions. Activity-dependent brain wiring abnormalities in shankopathies are important targets for therapeutic intervention, highlighting the need for further research in this area.

Reduced Expression of Hippocampal GluN2A-NMDAR Increases Seizure Susceptibility and Causes Deficits in Contextual Memory

Maria Florencia Acutain et al.

Summary: N-methyl-D-aspartate receptors consist of GluN1 obligatory subunits and regulatory subunits like GluN2A and GluN2B, whose expression is tightly regulated and has an impact on learning, memory, and pathologies. By reducing GluN2A expression, changes in GluN2A/GluN2B ratio were observed, leading to impaired memory and increased seizure susceptibility in rats. These findings suggest a role of GluN2A expression deregulation in hippocampal function and neurological disorders.

8p23.2-pter Microdeletions: Seven New Cases Narrowing the Candidate Region and Review of the Literature

Ilaria Catusi et al.

Summary: This study reported seven additional patients with 8p23.2-pter microdeletions, confirming the association of this deletion with clinical phenotypes such as cognitive impairment, developmental delay, and behavioral issues. The findings suggest that DLGAP2 may be the strongest candidate gene for neurodevelopmental/behavioral phenotypes associated with 8p23.2-pter microdeletions.

Hippocampal contributions to social and cognitive deficits in autism spectrum disorder

Sarah M. Banker et al.

Summary: Autism spectrum disorder (ASD) is characterized by impairments in social functioning as well as commonly impaired nonsocial cognition such as hippocampus-dependent spatial reasoning and episodic memory. The symptoms of ASD typically emerge between 12 and 24 months of age, a critical developmental period for the hippocampus. Despite evidence of hippocampal structural abnormalities in individuals with ASD, there has been relatively little focus on hippocampal function in human studies. This review highlights the hippocampus as a promising area of interest for future research in ASD.
