CCT2 is an aggrephagy receptor for clearance of solid protein aggregates

Xinyu Ma et al.

Summary: In this study, the researchers identified CCT2 as an autophagy receptor that regulates the clearance of aggregation-prone proteins in cells and mouse brains. CCT2 selectively associates with aggregation-prone proteins and interacts with autophagosome marker ATG8s. Furthermore, CCT2 specifically promotes the autophagic degradation of solid protein aggregates, independent of other ubiquitin-binding receptors or chaperone-mediated autophagy.

Nuclear miR-30b-5p suppresses TFEB-mediated lysosomal biogenesis and autophagy

Huijie Guo et al.

Summary: The study revealed that miR-30b-5p can inhibit TFEB-dependent gene transcription by binding to CLEAR elements in the nucleus, thus affecting lysosomal biogenesis and autophagy. Knocking out miR-30b-5p increases TFEB transactivation, leading to increased expression of autophagy and lysosomal biogenesis-related genes.

Annexin-A1 SUMOylation regulates microglial polarization after cerebral ischemia by modulating IKKα stability via selective autophagy

Xing Li et al.

Summary: The study demonstrates that SUMOylated ANXA1 promotes the beneficial phenotype polarization of microglia by suppressing NF-κB activation and proinflammatory mediators. SUMOylated ANXA1 targets the IKK complex to enhance IKKα degradation and facilitates the interaction between IKKα and NBR1 for selective autophagy-mediated degradation, leading to improved neurological function in cerebral ischemia. Up-regulation of ANXA1 SUMOylation in microglia may provide therapeutic benefits for cerebral ischemia.

Polyubiquitin and ubiquitin-like signals share common recognition sites on proteasomal subunit Rpn1

Andrew J. Boughton et al.

Summary: Proteasome-mediated substrate degradation relies on the coordinated actions of ubiquitin, UBL-containing shuttle proteins, and the proteasome. Research indicates that the proteasomal subunit Rpn1 associates with UBL-containing proteins and polyUb chains.

TRIP12 promotes small-molecule-induced degradation through K29/K48-branched ubiquitin chains

Ai Kaiho-Soma et al.

Summary: The study reveals that the ubiquitin ligase TRIP12 and K29/K48-branched ubiquitin chains play crucial roles in targeted protein degradation, accelerating the degradation process induced by PROTACs. This cooperative mechanism is unique and enhances the efficiency of degrading neo-substrates.

UBQLN2-HSP70 axis reduces poly-Gly-Ala aggregates and alleviates behavioral defects in the C9ORF72 animal model

Kejing Zhang et al.

Summary: UBQLN2 plays a critical role in reducing poly-GA aggregates and alleviating poly-GA-induced neurotoxicity in C9-ALS/FTD by recognizing HSP70 ubiquitination and promoting poly-GA degradation. Disruption of the interaction between UBQLN2 and HSP70 inhibits poly-GA aggregation in C9ALS/FTD iPSC-derived neurons. Enhancing HSP70 with the chemical compound 17AAG at the adult stage mitigates behavioral defects in poly-GA animals.
