Isolation and characterization of human embryonic stem cell-derived heart field-specific cardiomyocytes unravels new insights into their transcriptional and electrophysiological profiles

Arash Pezhouman et al.

Summary: In this study, we successfully isolated and characterized first and second heart field- and nodal-like cardiomyocytes from human embryonic stem cells using a double reporter line. Our findings provide a comprehensive understanding of the early development and functional properties of these different cardiomyocyte populations. We also identified two novel surface markers that can be used for purification of cardiomyocytes. This research has important implications for gene expression profiling, arrhythmia modeling, and drug screening, and it paves the way for the development of stem cell therapy for specific heart conditions.

Single-cell transcriptomic characterization of a gastrulating human embryo

Richard C. Tyser et al.

Summary: The single-cell transcriptional profile of a human embryo between 16 and 19 days after fertilization shows similarities and differences in gastrulation compared to mouse and non-human primate models. This study provides new insights into human development and offers valuable information for directed differentiation of human cells in vitro.

Retinoic acid signaling in heart development: Application in the differentiation of cardiovascular lineages from human pluripotent stem cells

Alexandra Wiesinger et al.

Summary: Retinoic acid signaling is crucial for heart development, influencing polarity establishment, progenitor formation, and ventricular wall growth. It is utilized in protocols for generating cardiac cell types from PSCs. Available protocols using RA for cardiovascular lineage differentiation and potential transcriptional mechanisms are discussed for further development of desired cardiac cell types from human PSCs.

Ventricular, atrial, and outflow tract heart progenitors arise from spatially and molecularly distinct regions of the primitive streak

Kenzo Ivanovitch et al.

Summary: The study reveals that the FHF and SHF in gastrulating mouse embryos are divided into distinct pools of progenitors at earlier stages than previously thought, each with a unique origin in the primitive streak. The findings also show that cardiac progenitors are prepatterned within the primitive streak, determining their allocation to different anatomical structures of the heart.

Generation of mature compact ventricular cardiomyocytes from human pluripotent stem cells

Shunsuke Funakoshi et al.

Summary: The study established a differentiation strategy for generating structurally, metabolically, and functionally mature compact ventricular cardiomyocytes from human pluripotent stem cells. These cardiomyocytes exhibited metabolic changes indicative of heart failure when challenged with pathological stimuli.
