The endocrine role of brown adipose tissue: An update on actors and actions

Aleix Gavalda-Navarro et al.

Summary: Brown adipose tissue (BAT) acts as an endocrine organ by secreting regulatory factors known as batokines, which exert effects on various tissues and organs including the liver, heart, and skeletal muscle. These batokines play a role in regulating insulin sensitivity, hepatic steatosis, glucose metabolism, and other physiological processes, and further research is needed to understand their impact on human metabolism.

Cardioprotective effect of NRG-4 gene expression on spontaneous hypertension rats and its mechanism through mediating the activation of ErbB signaling pathway

Zhang Yang et al.

Summary: This study aimed to explore the myocardial protective effect of Neuregulin-4 (NRG-4) gene expression on spontaneous hypertension (SHR) rats and its mechanism through mediating the activation of the ErbB signaling pathway. The results showed that upregulation of NRG-4 expression can inhibit cardiomyocyte proliferation and apoptosis, reverse myocardial fibrosis, and exert a cardioprotective role.

Summary: The prevalence of cardiomyopathy is higher in diabetic patients, yet there are no specific drugs for treating DCM despite decades of investigations. Targeting PPAR alpha activation may be an important strategy in patients with diabetes, metabolic syndrome, and atherosclerotic cardiovascular diseases.

Neuregulin 4 Downregulation Induces Insulin Resistance in 3T3-L1 Adipocytes through Inflammation and Autophagic Degradation of GLUT4 Vesicles

Francisco Diaz-Saez et al.

Summary: The downregulation of Nrg4 in adipocytes impairs insulin action, likely due to decreased insulin receptor and Glut4 proteins, while also increasing proinflammatory cytokine expression and upregulating proteins involved in autophagy. Restoring lysosomal activity can partially reverse these effects, indicating that Nrg4 plays a role in maintaining insulin responsiveness and regulating autophagy.

n-3 PUFAs protect against adiposity and fatty liver by promoting browning in postnatally overfed male rats: a role for NRG4

Fan Yang et al.

Summary: Early-life nutrition has a significant impact on adult metabolism. This study found that postnatal overnutrition increases the risk of NAFLD and decreases Nrg4 expression, while a postweaning n-3 PUFA diet enhances Nrg4 expression in adipose tissues and helps attenuate NAFLD induced by overnutrition. Additionally, external NRG4 reduces lipogenesis in hepatocytes and n-3 PUFAs may promote NRG4 production through the PPARG pathway.

Is there a role for neuregulin 4 in human nonalcoholic fatty liver disease?

Toon J. I. De Munck et al.

Summary: The study found no significant difference in plasma Nrg4 levels between NAFLD patients and healthy controls, and no correlation between plasma Nrg4 levels and hepatic fat fraction or presence of hepatic fibrosis in NAFLD patients. Additionally, gene expression profiles of the Nrg4-ErbB pathway in liver and VAT were not significantly different between NAFL, NASH, and obese controls.

Autophagy in the diabetic heart: A potential pharmacotherapeutic target in diabetic cardiomyopathy

Saikat Dewanjee et al.

Summary: Diabetes is clinically associated with a higher risk of cardiac failure, with diabetic cardiomyopathy being a common complication that results from myocardial infarction. The dysfunction of autophagy plays a crucial role in the development of diabetic cardiomyopathy, leading to impaired cellular homeostasis and potential pathogenic effects. Current research focuses on understanding the molecular mechanisms of autophagy regulation in diabetic cardiomyopathy and exploring pharmacotherapeutic strategies to target autophagy.

Preconditioning of mesenchymal stem cells with ghrelin exerts superior cardioprotection in aged heart through boosting mitochondrial function and autophagy flux

Liqiang Sun et al.

Summary: The study demonstrates that preconditioning MSCs with ghrelin can enhance cardioprotective effects, especially in elderly subjects, following IR injury. Preconditioned MSCs increase autophagy flux by downregulating Beclin-1 and P62 protein overexpression, and increasing LC3-II expression and the LC3-II/LC3-I ratio. Moreover, combining NMN with ghrelin-preconditioned MSCs synergistically enhances protective effects on infarct size and mitochondrial function.

Serum neuregulin 4 (NRG-4) level and non-alcoholic fatty liver disease (NAFLD): A case-control study

Helda Tutunchi et al.

Summary: This study found that patients with NAFLD had lower circulating NRG-4 levels compared to the control group. Individuals in the highest quartile of NRG-4 had lower BMI, WC, TG, and HOMA-IR than those in the lowest quartile. Multiple regression analysis showed BMI, WC, TG, HDL-C, and HOMA-IR were independently related to NRG-4 level. The odds of NAFLD decreased by 41% per 1 SD increase in NRG-4 level.
