Stereoselective O-Glycosylations by Pyrylium Salt Organocatalysis

Michael Martin Nielsen et al.

Summary: Despite being relatively inaccessible to non-specialists, the field of carbohydrate chemistry is aiming to increase the availability of stereoselective glycosylation chemistry, through the discovery of commercially available pyrylium salts as catalysts. These catalysts have shown to enable stereoselective O-glycosylations of a wide range of phenols and alkyl alcohols, providing better reproducibility for non-specialists. The reaction exhibits varying degrees of stereospecificity and likely proceeds via an S(N)2-like substitution mechanism.

J. Colin Mizia et al.

Summary: This study demonstrates the effectiveness of glycosyl tosylates for the synthesis of beta-glycosides of gluco-configured 2-deoxy sugars, and shows that the same method can be used for the selective synthesis of alpha-glycosides containing the allo-configured 2-deoxy sugar digitoxose.

N-Alkylated Analogues of Indolylthio Glycosides as Glycosyl Donors with Enhanced Activation Profile

Ganesh Shrestha et al.

Summary: In this study, the activation process of N-alkylated SInR derivatives was investigated by NMR, leading to the discovery of different activation pathways for SIn and SInR derivatives. The orthogonality between SInR leaving groups and thioglycosides, as well as the selective activation of thioimidates over SInR glycosides, were also examined.

An Efficient and Concise Synthesis of α-Galactosylceramide

Daniela Imperio et al.

Summary: A concise and stereoselective synthesis of alpha-GalCer was achieved by utilizing carefully designed protecting groups on both the donor and the acceptor to control the stereochemistry of the glycosylation reaction. This synthetic strategy proved to be efficient, giving good yields of alpha-GalCer in a small number of steps.

Summary: Vaccination is the most effective way to prevent invasive pneumococcal diseases, but current vaccines have limited coverage and new candidates are needed to control emerging serotypes. Research suggests that designing new saccharide fragments with shared structures among different serotypes could simplify vaccine composition and provide common antigen targets for protection.

Self-Promoted Glycosylation for the Synthesis of β-N-Glycosyl Sulfonyl Amides

Patrycja Mala et al.

Summary: N-Glycosyl N-sulfonyl amides were synthesized using self-promoted glycosylation without the need for catalysts or additives. The method allows for the selective beta-glycosylation of sulfonylated asparagine derivatives, providing access to glycosyl sulfonyl amides under mild conditions.

Exploring glycosyl sulphates as donors for chemical glycosylation

Daniela Imperio et al.

Summary: The preparation of anomeric tetrabutylammonium sulphates of glucose and galactose derivatives and their role as donors in glycosylation reactions were investigated in this study, with benzylated sulphates being more reactive than benzoylated donors. Metal triflates and basic conditions were shown to give good results under mild reaction conditions, suggesting a predominant S(N)2-like reaction mechanism.
