Summary: LNnT is a unique tetrasaccharide found in human milk, and is commercially added to infant formula as a functional fortifier. A novel LNnT-producing fl-1,4-GalT was identified from Aggregatibacter actinomycetemcomitans, showing high transglycosylation activity and low hydrolysis activity, making it a promising candidate for practical LNnT production.

Discovery and characterization of a novel α-L-fucosidase from the marine-derived Flavobacterium algicola and its application in 2′-fucosyllactose production

Summary: The study successfully synthesized 2'-fucosyllactose using the novel and efficient alpha-L-fucosidase OUC-Jdch16 from Flavobacterium algicola 12076, with a yield ranging from 84.82% to 92.15%. OUC-Jdch16 not only can be used for the preparation of 2'-FL, but also capable of transferring the fucosyl residue to other glycosyl receptors generating novel glycosylated compounds.

Engineered Glycosidases for the Synthesis of Analogs of Human Milk Oligosaccharides

Summary: Enzymatic synthesis is a biocompatible method for the synthesis of complex compounds, including human milk oligosaccharides. Previous studies have been limited to analytical reactions, but this study demonstrates the successful synthesis of three tetrasaccharides derived from lacto-N-tetraose using engineered glycosidases. The reaction products were isolated and characterized.

Sulfo-Fluorous Tagging Strategy for Site-Selective Enzymatic Glycosylation of para-Human Milk Oligosaccharides

Improvement of the Transglycosylation Efficiency of a Lacto-N-Biosidase from Bifidobacterium bifidum by Protein Engineering

Summary: In this study, the lacto-N-biosidase LnbB was engineered to efficiently synthesize lacto-N-tetraose (LNT) in one enzymatic step. The best variant for this purpose was found to be LnbB W394H, followed by W465H and Y419N, demonstrating high transglycosylation yield and regioselectivity. The optimization of substrate concentrations indicated that high initial substrate concentration and high acceptor-to-donor ratio are favorable for transglycosylation.
